摘  要:为探讨miR-146a基因单核苷酸rs2910164位点多态性与恩施地区痛风及高尿酸血症的相关性,分别收集健康对照者296例、原发性痛风患者159例和高尿酸血症患者197例。采用高单核苷酸多态性(Hi-single nucleotide polymorphism,Hi-SNP)结合多重PCR和高通量测序技术检测miR-146a基因rs2910164位点多态性,生化分析仪检测生化指标。采用SPSS17.0、Haplo View 4.2软件进行数据统计分析,用Logistic回归模型在95%CI内进行相关性风险度测算,分析miR-146a单核苷酸多态性rs2910164位点与原发性痛风和高尿酸血症的相关性。结果显示,rs2910164位点的基因频率在组间差异无统计学意义(P>0.05),GG基因型和G等位基因与痛风无相关性(OR=1.570,95%CI 0.710~3.470;OR=0.856,95%CI 0.650~1.128),与高尿酸血症亦无相关性(OR=0.937,95%CI 0.500~1.730;OR=0.819,95%CI 0.633~1.060)。该研究提示,miR-146a基因rs2910164位点单核苷酸多态性可能与恩施地区原发性痛风的发病及高尿酸血症无显著相关性。To investigate the possible association between miR-146 a single nucleotide ra2910164 polymorphism and gout/hyperuricemia in Enshi area,296 healthy controls,159 patients with primary gout and 197 patients with hyperuricemia were collected.Polymorphism of rs2910164 of miR-146a gene was detected by Hi-single nucleotide polymorphism(Hi-SNP)combined with multiple PCR and high-throughput sequencing technology.Biochemical indicators were determined by biochemical analyzer.The results showed that there was no statistical difference in rs2910164 gene frequency between the three groups,and both GG genotypes and G alleles were not susceptible to gout(OR=1.570,95%CI 0.710-3.470;OR=0.856,95%CI 0.650-1.128)and hyperuricemia(OR=0.937,95%CI 0.500-1.730;OR=0.819,95%CI 0.633-1.060).Taken together,single nucleotide polymorphism of miR-146 a gene rs2910164 is not associated with primary gout and hyperuricemia risk in Enshi area.
