摘 要:目的:探讨长链非编码RNA(LncRNA)FAS-AS1对胶质瘤细胞顺铂(Cis)耐药的影响及相关分子机制。方法:应用RT-PCR检测U87和U251细胞以及U87/Cis和U251/Cis细胞间差异表达LncRNA FAS-AS1;MTT法、CCK-8和流式细胞法检测过表达LncRNA FAS-AS1后的U87/Cis和U251/Cis细胞在一定浓度顺铂中的增殖能力、活力以及凋亡水平;免疫荧光分析顺铂作用下U87/Cis和U251/Cis细胞的自噬能力,并通过Western blot和RT-PCR进一步加以验证自噬及凋亡相关蛋白;随后进一步分析过表达LncRNA FAS-AS1后的U87/Cis和U251/Cis细胞其自噬能力的变化。结果:LncRNA FAS-AS1在U87/Cis和U251/Cis细胞系中显著低表达;在U87/Cis和U251/Cis细胞系中过表达LncRNA FAS-AS1,细胞的增殖能力无明显变化,细胞活力减弱,细胞凋亡水平显著增加;随着顺铂浓度的增加,U87/Cis及U251/Cis细胞的自噬及凋亡相关蛋白表达水平逐渐增加;过表达LncRNA FAS-AS1后U87/Cis及U251/Cis细胞的自噬相关蛋白表达水平降低。结论:LncRNA FAS-AS1通过抑制过度的自噬促进胶质瘤细胞对顺铂的敏感性。Objective:To investigate the effect of LncRNA FAS-AS1 on cisplatin(Cis)resistance of glioma cells and its related molecular mechanism.Methods:Differentially expressed LncRNA FAS-AS1 was detected by RT-PCR between U87 and U251 cells and between U87/Cis and U251/Cis cells.MTT assay,CCK-8 assay and flow cytometry assay were used to detect the proliferation,activity and apoptosis of U87/Cis and U251/Cis cells overexpressing LncRNA FAS-AS1 in a certain concentration of cisplatin.The autophagy capacity of U87/Cis and U251/Cis cells was analyzed by immunofluorescence,and the autophagy and apoptosis-related proteins were further verified by Western blot and RT-PCR.The changes of autophagy in U87/Cis and U251/Cis cells after over-expression of LncRNA FAS-AS1 were further analyzed.Results:The expression of LncRNAFAS-AS1 was significantly low in U87/Cis and U251/Cis cell lines.The over-expression of LncRNA FAS-AS1 in U87/Cis and U251/Cis cell lines resulted in no significant change in cell proliferation,but decreased cell viability and significantly increased cell apoptosis.With the increase of cisplatin concentration,the expression levels of autophagy and apoptosis-related proteins in U87/Cis and U251/Cis cells increased gradually.The expression levels of autophagy related proteins in U87/Cis and U251/Cis cells were decreased after the overexpression of LncRNA FAS-AS1.Conclusion:LncRNA FAS-AS1 promotes the sensitivity of glioma cells to cisplatin by inhibiting excessive autophagy.
