摘 要:目的了解海南省2019-2020监测年度乙型流感病毒Victoria系(BV)血凝素(hemagglutinin,HA)抗原性及其基因特征。方法按照时间不同从海南省5家流感监测网络实验室分离的流感毒株中选取14株BV系毒株,以WHO推荐的BV流感疫苗株B/Colorado/06/2017(2018-2020)为参考进行抗原性分析;对所有分离株进行一代全基因序列测定,应用生物信息学软件分析HA基因系统进化特征。结果与疫苗株相比,57.14%(8/14)的流感病毒分离株为类似株,42.86%(6/14)为低反应株;14株乙型流感病毒BV系分离株HA基因在进化上与疫苗株B/Colorado/06/2017亲缘关系较近,共同归属于1A分支,其中12株分离株聚为一支,以162-164位三缺失为分支特征。核苷酸和氨基酸同源性范围分别是98.6%~98.9%和98.3%~98.8%;14株BV流感分离株HA共有9个位点发生氨基酸变异:G129D、G133R、K136E、N197S、T199I、A202V以及162-164位缺失,涉及3个抗原表位。结论2019-2020监测年度海南省流行的乙型流感病毒分离株HA基因与疫苗株亲缘关系较近,但因HA氨基酸位点发生多处变异可能导致疫苗保护效果不佳。因此,需继续加强对流感流行情况和毒株型别及突变的监测。Objective To analyze the antigenicity and genetic characteristics of the hemagglutinin(HA)gene in the Victoria lineage of the influenza B virus(IBV)prevalent in Hainan Province from 2019-2020.Methods The materials used were 14 IBV strains isolated by the 5 influenza surveillance network laboratories in Hainan Province at different times,and their antigenicity was analyzed in comparison to that of the WHO recommended vaccine strain B/Colorado/06/2017(2018-2020).The genome sequences of the 14 isolated strains we’re obtained using Sanger sequencing.The phylogenetic characteristics of the HA gene were analyzed using bioinformatic software including DNAStar 7.0.1 and MEGA 7.0.14.Results Fifty-seven-point-one-four percent(8/14)of all influenza virus isolates were similar to the vaccine strain,and 42.86%(6/14)were less reactive strains.The HA gene of all 14 IBV strains was closely related to that of the vaccine strain B/Colorado/06/2017 in the evolutionary tree,and it belonged to the 1 A branch.Twelve strains clustered in a branch characterized by deletions at 162-164.The nucleotide similarity ranged from 98.6%to 98.9%and the amino acid similarity ranged from 98.3%to 98.8%.In addition,the HA gene of 14 IBV isolates had amino acid variations at nine sites:G129 D,G133 R,K136 E,N197 S,T199 I,A202 V,and deletions at 162-164.These variations involved three epitopes.Conclusion The HA gene of IBV isolates prevalent in Hainan Province from 2019-2020 were closely related to the vaccine strain.However,the accumulation of amino acid mutations in the HA gene may lead to influenza vaccines providing poor protection.Therefore,attention should be paid to continued enhancement of the monitoring of the prevalence of influenza,the virus type,and mutations.
