摘  要:目的:研究骨碎补总黄酮对牵张成骨模型大鼠成骨相关蛋白表达的影响。方法:选用SPF级健康雄性SD大鼠60只,随机分为模型组、低剂量组、中剂量组、高剂量组,每组15只,构建左侧胫骨牵张成骨模型,经5天静止期,使用胫骨环状外固定器以0.2 mm/d的速度牵引,连续20天。术后第1天起,低、中、高剂量组予骨碎补总黄酮浓度67、201、603 mg/kg行灌胃处理,模型组予等量生理盐水,灌胃30天后处死动物,留取标本,行Western Blotting检测成骨生长相关蛋白BMP-2、CTGF、FGF-2的表达。结果:与模型组比较,中药组成骨相关蛋白BMP-2、CTGF、FGF-2的表达均明显提高,差异有统计学意义(P﹤0.05),其中以高剂量组效果最明显,具体表现为CTGF的相对表达量提高3.4倍,BMP-2提高5.125倍,FGF-2提高6.333倍。结论:骨碎补总黄酮可通过促进DO大鼠成骨相关蛋白BMP-2、CTGF、FGF-2的表达加速新生骨形成及软组织修复。Objective:To observe the effect of osteopractic total flavone on expressions of osteogenic associated proteins in model rats with distraction osteogenesis.Methods:60 healthy male SD rats of SPF grade were selected and randomly divided into model group,low-,medium-and high-dose group,15 rats in each group.The model of left tibial distraction osteogenesis was established.After 5 days of stationary phase,the established model was drawn by tibial annular external fixator at the speed of 0.2 mm/d continuously for 20 days.From the first day after operation,low-,mediumand high-dose group were administered osteopractic total flavone whose concentration was respectively 67,201and 603 mg/kg by gavage,while the model group was administered same amount of normal saline.After gavage for 30days,the rats were killed,and the samples were taken.The expressions of osteogenic growth associated proteins such as BMP-2,CTGF and FGF-2 were detected by WesternBlotting.Results:Compared with model group,the expressions of osteogenic associated proteins BMP-2,CTGF and FGF-2 in TCM group were significantly increased,and the difference was statistically significant(P﹤0.05).Among them,the effect of high-dose group was the most obvious.The specific performance was that the relative expression of CTGF was increased by 3.4 times,the relative expression of BMP-2 by 5.125 times and the relative expression of FGF-2 by 6.333 times.Conclusion:Osteopractic total flavonoid can accelerate the osteogenesis and repair of soft tissue by promoting the expressions of osteogenesis related proteins BMP-2,CTGF and FGF-2 in DO rats.
