摘 要:[目的]研究将生长/分化因子-10(GDF10)作为候选基因,探讨其多态与不同性别的德州驴生长性状之间的关系。[方法]通过对440头德州驴进行多态检测,鉴定得到3个SNPs位点,并对SNP位点进行分型。利用SPSS 18.0软件分析3个SNPs位点的不同基因型与德州驴9个生长性状的关联分析。[结果]结果显示,3个位点均处于Hardy-Weinberg平衡状态(P>0.05);群体遗传参数分析发现,GDF10基因的SNP位点的纯合度较高,多态信息含量均处于中度多态(0.25G位点AA基因型个体的尻高显著高于GA基因型个体(P<0.05);g.2041898A>G位点AA基因型个体的体高和尻长显著高于GA基因型个体(P<0.05);g.2043035 C>T位点上CC基因型个体的尻长显著高于CT基因型个体(P<0.05)。在公驴中,g.2043035 C>T位点CC基因型个体的体高显著低于CT基因型(P<0.05)。[结论]综上所述,GDF10基因多态性与德州驴的部分生长性状相关,有潜力作为德州驴的生长发育性状分子标记。[Objective] In this study, growth/differentiation factor-10(GDF10) was selected as a candidate gene to explore the association between GDF10 polymorphism and growth traits of Dezhou donkeys of different genders. [Method] In 440 Dezhou donkeys, 3 SNPs were detected, and were genotyped. SPSS 18.0 software was used to analyze the association between three genotypes at different SNP loci and nine growth traits of Dezhou donkeys. [Result] The results showed that the three loci were in hardy-Weinberg equilibrium(P> 0.05). Analysis of population genetic parameters showed that there is a high degree of homogeneity in the polymorphic loci of GDF10 gene, and the content of polymorphic information was moderate(0.25< PIC< 0.50). The results of association analysis showed that in jennies, the rump high of AA genotype was significantly higher than that of GA genotype at g.2031481 A>G locus(P<0.05), at g.2041898 A>G locus, the body height and rump length of AA genotype were significantly higher than those of GA genotype(P<0.05), and at g.2043035 C>T locus, the rump length of CC genotype was significantly higher than that of CT genotype(P<0.05). In jackasses, the body height of CC genotype at the g.2043035 C>T locus was significantly lower than that of CT genotype(P<0.05). [Conclusion] In conclusion, GDF10 gene polymorphism is related to the growth traits of Dezhou donkeys and can be used as a molecular marker for the growth and development of Dezhou donkeys.
