摘 要:研究刺五加多糖(ASPS)对免疫抑制雏鸡黏膜淋巴组织损伤的修复作用,从器官和组织水平对ASPS的免疫增强作用进行评价。将1日龄海兰褐公雏饲养至4日龄时选取300只,随机分为3组,每组100只,第1组为空白对照组,第2组为环磷酰胺(CTX)致免疫抑制组,第3组为(ASPS+CTX)组。第2组和第3组均按照80 mg/(kg·d)的剂量皮下注射CTX,连续注射3 d。从10日龄开始,第3组每天每只鸡皮下注射200 mg/mL ASPS 0.2 m L,连续注射3 d。第1组和第2组以同样方式注射等量无菌生理盐水。注射后7 d、14 d、21 d和28 d,分别取盲肠扁桃体(CT)、肺脏和眼结膜,通过H.E染色对黏膜淋巴组织的组织结构变化进行观察。结果表明,CTX能够造成CT的黏膜固有层组织萎缩和生发中心显著减少或消失、肺脏内支气管相关淋巴组织(BALT)萎缩或消失以及结膜相关淋巴组织(CALT)萎缩形成纤维结缔组织,从而显著抑制了黏膜淋巴组织的发育。在ASPS的作用下,CTX所导致的CT、BALT和CALT组织结构损伤均得到显著改善。与CTX组比较,从免疫后14 d开始,(ASPS+CTX)组CT黏膜固有层中生发中心开始出现并增多,而且轮廓逐渐清晰;免疫后21 d和28 d时,BALT较CTX组显著增大,而且与正常组相近;各日龄(ASPS+CTX)组CALT面积较同日龄CTX组明显增加,但是淋巴组织中并未出现生发中心。因此,ASPS对CTX造成的CALT组织结构损伤修复效果低于对CT和BALT的修复效果。结果提示,ASPS能够拮抗CTX对雏鸡黏膜淋巴组织的抑制作用,修复CTX造成的各种黏膜淋巴组织的结构损伤,从而进一步增强黏膜免疫功能,证明ASPS可以作为理想的黏膜免疫增强剂使用。This experiment was conducted to study the ameliorative effect of Acanthopanax senticosus polysaccharide(ASPS)on morphologic injury of mucosal lymphoid tissue in immunosuppressive chickens,and to evaluate the immune-enhancing effect of ASPS at the organ and tissue level.A total of 3004-day-old Hy-Line brown male chicks were randomly divided into 3 groups with 100 chickens in each group.The first group was the blank control group,the second group was the cyclophosphamide(CTX)-induced immunosuppression group,and the third group was the ASPS+CTX group.Group 2 and group 3 were subcutaneously injected CTX at a dose of 80 mg·kg-1 daily for3 consecutive days.From 10-day-old,group 3 was subcutaneously injected with 0.2 mL ASPS(200 mg·mL;)per chicken every day for 3 consecutive days,and group 1 and group 2 were injected with the same amount of sterilized normal saline in the same way.Cecal tonsil(CT),lung and eye conjunctiva were taken at 7 d,14 d,21 d and 28 d after injection,and histological changes of mucosal lymphoid tissue were observed by H.E staining.The results showed that CTX could significantly inhibit the development of mucosal lymphoid tissue by causing atrophy of lamina lamina and germinal center,atrophy or disappearance of bronchial associated lymphoid tissue(BALT)and atrophy of conjunctival associated lymphoid tissue(CALT).Under the effect of ASPS,CT,BALT and CALT morphologic injury induced by CTX were significantly improved.Compared with the CTX group,from 14 d after immunization,germinal centers in the CT mucosal lamina propria of the ASPS+CTX group began to appear and increase,and the contour was gradually clear.At 21 d and 28 d after immunization,BALT significantly increased compared with CTX group,and was similar to normal group.The CALT area of ASPS+CTX group at each day of age was significantly higher than that of CTX group at the same day of age,but there was no germinal center in the lymphoid tissue.Therefore,the repair effect of ASPS on the structural damage of CALT in CTX group was lower than t
