摘 要:研究叶下珠醇提物对攻毒肠炎沙门菌(Salmonella enteritidis)毒素小鼠血清、肝、脾和空肠炎症因子水平的影响。将50只昆明小鼠随机分为空白对照组(CK)、肠炎沙门菌毒素组、叶下珠醇提物高剂量组、中剂量组和低剂量组(40 mg/mL、20 mg/mL和10 mg/mL)。各组灌胃叶下珠醇提物7 d,除对照组外,其余各组均腹腔注射肠炎沙门菌毒素,24 h后采血,扑杀各组小鼠,快速分离肝、脾和空肠组织,ELISA测定小鼠血清中IL-6、TNF-α和IL;0水平。结果表明,与对照组相比,肠炎沙门菌毒素组小鼠腹泻等临床症状明显,剖检可见肝、脾、肠出血肿胀等病变,叶下珠醇提物各剂量组小鼠临床症状和脏器损伤均明显轻于肠炎沙门菌毒素组。炎症因子检测结果显示,与对照组相比,肠炎沙门菌毒素组血清中IL-6水平显著下降(P<0.05),但TNF-α和IL;0水平变化不显著(P>0.05),肝、脾和空肠中IL-6、TNF-α水平均极显著升高(P<0.01),IL;0水平均极显著降低(P<0.01)。叶下珠醇提物各剂量组血清、肝、脾、空肠中IL-6和TNF-α水平显著降低,IL;0水平显著升高,综合评价以低剂量组效果最佳。提示叶下珠醇提物能通过抑制炎症反应,拮抗肠炎沙门菌毒素对小鼠的损伤。To study the effects of ethanol extract of Phyllanthus urinaria on the levels of inflammatory factors in serum,liver,spleen and jejunum of mice challenged with Salmonella enteritidis toxin,50 Kunming mice were randomly divided into blank control group(CK),toxin group,Phyllanthus urinaria alcohol extract high(PH),medium(PM)and low(PL)dose groups(40 mg·mL;,20 mg·mL;and 10 mg·mL;). After 7 days of intragastric administration of ethanol extract of Phyllanthus urinaria,except for the control group,all mice were intraperitoneally injected with toxin. 24 h later,the eyeballs of mice were taken for blood collection to kill the mice in each group. The liver,spleen and jejunum tissues were quickly separated,and the levels of IL-6,TNF-α and IL;0 in serum were determined by ELISA. The results showed that compared with the control group,diarrhea and other clinical symptoms were obvious in toxin group,liver,spleen,intestinal bleeding,swelling and other pathological changes could be seen in autopsy,and the clinical symptoms and visceral injury in each dose group of Phyllanthus urinaria were significantly lower than those in toxin group. Compared with the control group,the level of IL-6 in toxin group was significantly decreased(P<0.05),but the level of TNF-α in toxin group had no significant difference(P>0.05). The levels of IL-6 and TNF-α in liver,spleen and jejunum were extremely significant increased(P<0.01),and the levels of IL;0 were extremely significant decreased(P<0.01). All dose groups could effectively decrease the levels of IL-6 and TNF-α and increase the levels of IL;0 in serum,liver,spleen and jejunum. Comprehensive evaluation showed that PL group was the best. The results showed that the ethanol extract of Phyllanthus urinaria could antagonize the damage of Salmonella enteritidis toxin in mice by inhibiting the inflammatory reaction. It could be used as a potential green substitute for anti-inflammatory drugs and had a broad development and application prospect.
