摘  要:磷脂酰乙醇胺结合蛋白(phosphatidyl ethanolamine-binding protein,PEBP)基因家族的氨基酸结构中有保守的磷脂酰乙醇胺结合蛋白结构域,对控制植物花器发育和开花时间具有重要作用。本试验旨在研究CsPEBP基因家族对甜橙花期诱导的调控模式,利用PEBP蛋白特征结构域PBP的隐马尔可夫模型在甜橙基因组中进行HMMER分析,鉴定出7个Cs PEBP基因,并对CsPEBPs编码蛋白的理化性质、蛋白基序、进化关系、顺式调控元件和表达谱等方面进行研究。结果表明CsPEBPs分布在甜橙5条染色体上。遗传进化树聚类成4个亚族,CsPEBP基因家族成员被分在TFL1-subclade、MFT-subclade、FT-subclade 3个亚族中。7个CsPEBP基因均含有光响应元件,表明CsPEBPs受光诱导调控。实时荧光定量PCR(RT-qPCR)分析发现,CsPEBP基因在两个甜橙品种花芽和叶片的表达模式不同,其中CsFT1和CsFT2在两甜橙品种花芽和叶片中的相对表达量均有显著上调的趋势,而CsTFL1a、CsTFL1b和CsTFL1c在它们花芽和叶片中的相对表达量均显著下调;另外,早熟品种‘赣南早’开花时期,‘赣南早’花芽和叶片中CsFT亚族基因的表达量高于对照品种‘纽荷尔’,而CsTFL1亚族基因是相反的,说明花芽细胞内CsFTs高表达和CsTFL1s低表达导致了甜橙花期提前,Cs PEBPs对调控甜橙开花时间起重要作用。本研究获取了甜橙PEBP基因家族成员的序列和部分功能信息,为进一步分析CsPEBP基因家族在甜橙成花转变中的功能提供参考。The amino acid structure of Phosphatidyl Ethanolamine-Binding Protein(PEBP)gene family has a conserved Phosphatidyl Ethanolamine Binding Protein(PEBP)domain,which plays an important role in controlling plant floral organ development and flowering time.This study was conducted to study the regulation mode of CsPEBP gene family on flowering induction of Citrus sinensis.Seven CsPEBP genes were identified by HMMER analysis using the hidden Markov model of PEBP characteristic domain PBP in the whole genome of Citrus sinensis.And the physicochemical properties,protein motif,evolutionary relationship,cis-acting elements and expression profile of CsPEBP encoded protein were studied.The results showed that CsPEBPs were distributed on five chromosomes of Citrus sinensis.The genetic evolutionary tree was clustered into four subgroups,and the members of CsPEBP gene family were divided into three subgroups:TFL1 subclade,MFT subclade and FT subclade.All Cs PEBP genes contain light response elements,indicating that CsPEBPs are regulated by light induction.Real time fluorescence quantitative PCR(RT-qPCR)analysis showed that the expression patterns of CsPEBP genes in flower buds and leaves of two Citrus sinensis varieties were different.The relative expressions of CsFT1 and CsFT2 in the flower buds and leaves of the two Citrus sinensis varieties were significantly up-regulated,while the relative expressions of CsFT1a,CsTFL1b and CsTFL1c in their flower buds and leaves were significantly down regulated.In addition,during the flowering period of early maturing variety’Gannan Zao’,the expression of CsFT subfamily genes in flower buds and leaves of’Gannan Zao’is higher than that of control variety’Newhall’,while the expression of CsTFL1subfamily genes was the opposite,indicating that the high expression of Cs FTs and low expression of CsTFL1s in flower bud cells lead to the early flowering period of Citrus sinensis,and CsPEBPs plays an important role in regulating the flowering time of Citrus sinensis.In this study,the se
