摘 要:稻瘟病菌(Magnaporthe oryzae)引起的水稻稻瘟病,每年造成全球水稻减产约10%~30%,对全球粮食生产构成严重威胁。MOB(Mps one binder)蛋白家族成员在物种进化过程中高度保守,并作为重要激酶调控因子与NDR(Nuclear DBF2-related)蛋白激酶等结合,在细胞周期的有丝分裂退出和胞质分离等事件中发挥重要作用。本研究利用功能丧失和获得的方法建立了MoMOB1敲除突变体,对突变体进行了功能回补;并对背景菌株、敲除突变体和回补菌株,在形态特征和致病性等方面进行了生物性状分析,以解析稻瘟病菌中MoMOB1的生物学功能。与背景菌株Ku80相比,MoMOB1缺失后稻瘟病菌生长速率明显减慢,分生孢子萌发延迟,突变体产孢量显著降低,分生孢子出现畸形且隔膜数目异常,部分分生孢子没有隔膜或只有一个隔膜。致病性分析发现,ΔMoMOB1突变体致病性明显减弱。回补菌株的表型与背景菌株Ku80相似。综上所述,MoMOB1在稻瘟病菌营养生长、产孢量、分生孢子发育、附着胞形成和致病性方面发挥关键作用。Rice blast,caused by Magnaporthe oryzae,reduces global rice yield about 10%~30%annually.It is a serious threat to rice production all over the world.Members of the MOB(Mps one binder)protein family are highly conserved in species evolution process.MOB1 as key kinase regulators bind to NDR(Nuclear DBF2-related)protein kinases playing an important role in critical events such as mitotic exit and cytokinesis of cell cycle.In order to elucidate biological function of MoMOB1 in M.oryzae,MoMOB1 gene knock-out mutant and complementary strain were established by loss-and gain-of-function approaches.The strains were analyzed in morphological characteristics and pathogenicity along with background strain Ku80.Comparing to Ku80,the growth rate ofΔMoMOB1 is significantly slowed down and conidia germination lagged.The mutant has a sharp decrease in conidiation and conidia deformity with abnormal septum.Some of the conidia has only one septum or no septum.ΔMoMOB1 mutant decreased in pathogenesity by assays.The phenotype ofΔMoMOB1-Com complementation was similar to that of the background strain Ku80.Taken together,MoMOB1 plays a key role in the vegetative growth,conidiation,conidium development,appressorium formation and pathogenesis in M.oryzae.
