摘  要:紫薇(Lagerstroemia indica)是中国及世界上重要的木本观花植物,高效的遗传转化技术是开展紫薇基因功能验证和分子育种的关键技术之一。因此,为构建紫薇高效的遗传转化技术体系,本研究以紫薇腋芽为受体材料,探究了卡那霉素浓度、农杆菌菌液浓度和侵染时间对紫薇遗传转化效率的影响。结果表明,腋芽萌发筛选最适的卡那霉素浓度为300 mg/L,最优转化条件为农杆菌菌液浓度OD_(600)=0.2~0.8、侵染时间10~15 min,遗传转化效率最高为25.93%。预培养、共培养及筛选培养的培养基为MS+1.0 mg/L 6-BA+0.1 mg/L NAA+30 g/L Sugar。本研究建立了不依赖愈伤组织的紫薇高效遗传转化技术体系,为紫薇重要性状基因的功能验证及分子育种提供了重要工具。Crape myrtle(Lagerstroemia indica)is an important woody ornamental plant in the world.High-efficiency genetic transformation technology is one of the key technologies for the gene function verification and molecular breeding in crape myrtle.In order to construct an efficient genetic transformation technology system for crape myrtle,axillary buds were used as recipient material in this study,and the effects of kanamycin concentration,Agrobacterium liquid concentration and infection time on the genetic transformation efficiency were explored.The results showed that the optimal kanamycin concentration was 300 mg/L.The optimal transformation conditions were:Agrobacterium concentration of OD_(600)=0.2~0.8,infection for 10~15 min.The highest transformation efficiency was 25.93%.The common medium for pre-,co-and selection-culture was MS+1.0 mg/L 6-BA+0.1 mg/L NAA+30 g/L sugar.In this study,a highly efficient genetic transformation technology system that does not rely on callus was established,which could provide an important tool for gene functional verification of important traits and molecular breeding in Lagerstroemia indica.
