摘 要:为了解牡丹远缘杂交组合遗传差异,筛选优势杂交材料。本研究综合利用形态分析和目标起始密码子多态性分子标记(start codon targeted polymorphism,SCoT)技术对远缘杂交亲本紫牡丹、‘珊瑚台’及其4个杂交后代品种进行形态性状和遗传差异分析。结果表明,形态学分析显示杂交后代较多地遗传了父本‘珊瑚台’的性状特征,形态上表现为花径增大、重瓣化、雄蕊瓣化等特征,后代的性状更加优良,观赏价值更高,杂种优势明显。SCoT分子标记对该杂交组合及其后代品种扩增结果显示其多态性较好,证明该分子标记能有效用于牡丹遗传多样性分析应用。利用NTSYS-pc 2.10e软件分析了6份牡丹材料的遗传相似系数(GS),变异范围在0.2647~0.7059之间,在GS值为0.496水平上聚为3个大类,其中杂交后代‘红珊瑚’和‘紫珊瑚’之间的遗传相似系数最大,为0.706,而母本紫牡丹和后代品种‘紫珊瑚’之间的遗传相似系数最小,为0.265。因此,综合运用形态学和分子标记分析方法,能有效地揭示远缘杂交亲本及后代间的遗传差异大小,可为牡丹的新品种培育提供参考。In ord er to understand the genetic differences of distant hybrid combinations of peony,screen the dominant hybrid materials.The morphological characters and genetic differences of the distant hybrid parents(Paeonia delavayi and’Shanhutai’)and their four progenies were analyzed by morphological analysis and start codon targeted polymorphism(SCoT)molecular markers.The results showed that the morphological analysis showed that the hybrid progeny had inherited the characteristics of the original variety’Shanhutai’of the male parent,and showed the excellent characteristics of enlarged flower diameter,double petalization and stamen petalization,and the progeny’s characteristics were better,the ornamental value was higher,and showed strong heterosis.SCoT molecular markers were used to amplify the hybrids and their progenies.The results showed that the markers were effective in the analysis of peony polymorphism.The genetic similarity coefficient(GS)of 6 peony materials was analyzed by NTSYS-pc 2.10e software.The variation range was 0.2647~0.7059,and it can be divided into three categories at GS value of 0.496.The genetic similarity coefficient between’Hongshanhu’and’Zishanhu’was the largest(0.706),while that of Paeonia delavayi and’Zishanhu’was the smallest,which is 0.265.Therefore,the comprehensive use of morphological and molecular marker analysis methods can effectively reveal the genetic differences between distant hybrid parents and progenies,and provide a reference for the cultivation of new varieties of peony.
