摘 要:目的观察灯盏乙素对结肠癌HT-29细胞株增殖、凋亡、转移影响与circ-ACVR2A表达的相关性。方法将结肠癌HT-29细胞株分为阴性对照组,灯盏乙素10、20、40μmol·L^(-1)组,采用CCK-8法检测细胞增殖情况,AnnexinV-FITC/propidium iodide(PI)流式细胞仪检测细胞凋亡情况,Western blot检测Bcl-2、Bax蛋白表达及Bcl-2/Bax比值,Transwell实验检测细胞侵袭情况,qRT-PCR检测circ-ACVR2A相对表达量。结果与阴性对照组比较,灯盏乙素各剂量组处理后24、48和72 h,HT-29细胞增殖出现明显下降,细胞株生长抑制率显著升高,有统计学意义(P<0.05)。与阴性对照组比较,灯盏乙素10μmol·L^(-1)组凋亡细胞率为11.3%,灯盏乙素20μmol·L^(-1)组凋亡细胞率为17.1%,灯盏乙素40μmol·L^(-1)组凋亡细胞率为31.2%,抑制Bcl-2表达,促进Bax表达,Bcl-2/Bax比值降低,细胞侵袭数量明显下降,circ-ACVR2A相对表达量增加,差异具有统计学意义(P<0.05)。结论灯盏乙素能明显抑制结肠癌细胞株增殖与侵袭,促进细胞凋亡,其机制可能与降低Bcl-2/Bax比值、增加circ-ACVR2A表达有关。Objective To observe the effects of scutellarin on the proliferation,apoptosis and invasion of colon cancer HT-29 cell line via regulating the circ-ACVR2 A expression.Methods HT-29 cell lines were divided into the control group and scutellarin groups(10,20,40μmol·L^(-1)).Cell proliferation was detected by CCK-8,and the apoptosis was measured by flow cytometry with annexinv-FITC/PI staining.Bcl-2 and Bax protein expression were detected by Western blot,the cell invasion was detected by transwell assay,and relative expression of circ-ACVR2 A was detected by qRT-PCR.Results Compared with the control group,the proliferation of HT-29 cells decreased and the inhibition rate of cell line increased significantly in the scutellarin group after treatment with scutellarin(10,20,40μmol·L^(-1))for 24,48 and 72 h(P<0.05).Compared with the control group,the apoptotic cell rates of 10μmol·L^(-1),20μmol·L^(-1)and 40μmol·L^(-1)scutellarin groups were 11.3%,17.1%and 31.2%,respectively.For scutellarin groups,Bcl-2 expression was inhibited while Bax expression was promoted;the number of cell invasion decreased and the relative expression of circ-ACVR2 A increased significantly(P<0.05).Conclusion Scutellarin significantly inhibits colon cancer proliferation,invasion and promotes apoptosis.The mechanism may be related to the decrease of Bcl-2/Bax ratio and the increase of circ-ACVR2 A expression.
