摘 要:小西葫芦黄花叶病毒(zucchini yellow mosaic virus,ZYMV)严重危害西瓜等葫芦科作物。西瓜938-16-B是对ZYMV产生过敏性坏死反应(HR)的抗性自交系。为研究其对ZYMV的抗性遗传特征,以其为父本与感病亲本H_(1)杂交,构建了包含310个单株的F_(2)群体及BC_(1)群体,通过对亲本、F_(1)、BC1和F_(2)的抗病性鉴定,分析938-16-B的抗性遗传规律。结果发现:温度对西瓜幼苗感染ZYMV后HR的形成有一定影响,低温(<28℃)下比高温(>30℃)更易产生HR反应。在严格控制温度(23~28℃)的条件下对杂交后代群体进行接种抗病性鉴定,结果显示,F_(2)群体的抗、感分离比符合3︰1(χ^(2)=0.520,P=0.470);BC1的抗、感比例符合1︰1(χ^(2)=0.040,P=0.841),而亲本938-16-B和F_(1)全部抗病,亲本H_(1)全部感病,表明西瓜938-16-B抗ZYMV为单基因显性遗传。Zucchini yellow mosaic virus is a serious emerging virus that has threatened the economically important cucurbits industry in major production areas worldwide.Watermelon inbred line 938-16-B had been identified as resistance to ZYMV in our previous work.It is characterized by hypersensitive response(HR)after infection by ZYMV.Here,we investigated the inheritance of resistance to ZYMV in 938-16-B.In this study,F_(2)generation with 310 individuals and BC_(1)generation were generated through crossing 938-16-B with ZYMV-susceptible inbred line H_(1).According to the segregation ratio of resistance in BC_(1)and F_(2)generation,ZYMV resistance in 938-16-B would be established.During the process of inoculation,we found hypersensitive response(HR)would not be induced when the temperature was over 30℃,which suggested the resistant gene was temperature-sensitive.Therefore,identification of resistance in the F_(2)and BC_(1)population was carried out under the condition of 23–28℃.Results showed that,the segregation ratios of resistance to susceptibility in F_(2)and BC_(1)were 3︰1 and 1︰1,respectively,which revealed the resistance to ZYMV in watermelon is controlled by a single dominant gene.
