摘 要:为建立金银木松散型胚性愈伤组织培养体系,该文研究金银木不同外植体、不同植物激素配比对金银木松散型胚性愈伤组织诱导与继代保持的影响。结果表明,金银木叶片为诱导愈伤组织最适外植体;叶片在MS(Murashige&Skoog Basal Medium,MS基础培养基)+0.5 mg/L BA(Benzylaminopurine,苄氨基腺嘌呤)+1.0 mg/L 2,4-D(2,4-Dichlorophenoxyacetic acid,2,4-二氯苯氧乙酸,)培养基上可诱导出松散型愈伤组织,愈伤组织诱导率达100%;将松散型愈伤组织转接到MS+0.5 mg/L BA+0.25 mg/L 2,4-D培养基上,每20 d继代培养一次,继代培养2~3次后可获得金银木松散型胚性愈伤组织。In order to establish the culture system of loose embryogenic callus of Lonicera maackii,the effects of different explants and different plant hormone ratio on the induction and subculture maintenance of loose embryogenic callus were studied.The results showed that the leaf of L.maackii was the most suitable explant for callus induction;loose callus could be induced from the leaf of L.maackii on the medium of MS(Murashige&Skoog Basal Medium)+0.5 mg/L BA(Benzylaminopurine)+1.0 mg/L 2,4-D(2,4-Dichlorophenoxyacetic acid),and the callus induction rate reached 100%;the loose callus were transferred to the medium of MS+0.5 mg/L BA+0.25 mg/L 2,4-D,and subcultured every 20 days,the loose embryogenic callus were obtained after subculture for 2-3 times.
