摘 要:目的在研究孕酮(progesterone)对脊髓外伤(Spinal Cord Injury,SCI)的治疗作用中,通过检测孕酮治疗后致伤脊髓组织中的c-myc的表达情况及可能治疗机制。方法36只成年SD大鼠随机分为假手术组(12只)、动物模型组(12只)及治疗组(12只),通过改良Allen’s法制作SCI模型,治疗组予孕酮治疗7 d,观察记录各组大鼠伤后行为学评分(BBB评分)、磁共振表现及脊髓节段病理表现,检测c-myc基因及蛋白的表达。结果治疗组在BBB评分上较模型组明显升高,局部神经坏死程度及水肿程度减轻;c-myc基因及蛋白表达明显减少(P<0.05)。结论孕酮对SCI具有短期积极治疗作用,其主要机制可能与抑制c-myc表达有关。Objective In the study of the therapeutic effect of progesterone(PG)on spinal cord injury(SCI),the partial mechanism was explored by detecting the expression of c-myc in the injured spinal cord tissue after PG treatment.Methods 36 adult Sprague-Dawley rats were randomly divided into the sham(12 rats)、animal model group(12 rats)and the treatment group(12 rats).The SCI animal model was established by improved Allen’s and was given a treatment with PG for 7 days.The ethological scores(BBB scores)、MRI and the pathological manifestations of spinal cord segments of the rats in each group were observed and recorded,and the expression of c-myc was detect by PCR and Western blot.Results The BBB scores in the treatment group was significantly increased,and the degree of local nerve necrosis was slightly less.The expression of c-myc was significantly decreased(P<0.05).Conclusion PG has a positive effect on SCI,and its mechanism may be related to the inhibition of c-myc.
