中药苏木抑制胃癌活性组分鉴定及抑制效果研究

摘　　要：为了筛选中药苏木中抑制胃癌的活性成分,并探讨其对胃癌细胞BGC-823及荷BGC-823胃癌裸鼠的抑制作用,采用不同提取方法制备10种苏木提取物样品,用高效液相色谱法分别测定其组分含量;用MTT法检测其对BGC-823细胞的增殖抑制效果,通过分析各提取物主要组分的含量变化与其对BGC-823细胞抑制率的相关性筛选其抗癌活性成分。选择抗癌活性成分含量最高的苏木提取物为实验样品,研究其在不同作用浓度和作用时间下对BGC-823细胞的抑制效果及对荷BGC-823胃癌裸鼠的抑制效果。结果表明,苏木提取物能显著抑制BGC-823细胞的增殖,巴西苏木素是其抑癌主要活性成分;苏木提取物样品中巴西苏木素含量最高达55.74%,其对BGC-823细胞的抑制作用具有剂量和时间依赖性;对荷BGC-823胃癌裸鼠有很好的肿瘤抑制作用,中剂量组抑瘤率为48.55%,高剂量组抑瘤率为60.86%。To screen the active components of Sappan L.for inhibiting gastric cancer,and to investigate its inhibitory effect on gastric cancer cells BGC-823 and nude mice bearing BGC-823 gastric cancer,10 samples extracted from Sappan L.were prepared by different extraction methods,and their component contents were determined by high performance liquid chromatography(HPLC),and their proliferation inhibitory effect on BGC-823 cells was detected by MTT method.The anti-cancer active ingredients were screened by analyzing the correlation between the content changes of the main components in each extract and its inhibition rate on BGC-823 cells.The extract from Sappan L.with the highest content of anti-cancer active ingredients was selected as the experimental sample,its inhibitory effect on BGC-823 cells and the inhibitory effect on BGC-823 gastric cancer bearing nude mice at different concentration and time were studied,respectively.The results indicate that the extracts from Sappan L.can significantly inhibit the proliferation of BGC-823 cells whose main anti-cancer active ingredient is Brazilin.The Brazilin content in extract samples from Sappan L.is up to 55.74%,which has a dose-and time-dependent effect on the inhibition of BGC-823 cells and has a great anti-tumor effect on nude mice bearing BGC-823 gastric cancer,and the tumor inhibition rate in the middle-dose group is 48.55%,and that in the high-dose group is 60.86%.
