摘 要:目的探讨小檗碱对利血平诱导的抑郁症模型小鼠行为学变化的影响。方法将40只实验动物随机分为正常对照组、模型组、小檗碱组及氟西汀组,每组各10只。模型组、小檗碱组及氟西汀组小鼠每日腹腔注射利血平200 mg/kg(生理盐水配制),正常对照组每日腹腔注射等量的生理盐水,每组小鼠连续注射4周。造模4周后采用灌胃法给药,每天1次,共21 d。小檗碱组190 mg/kg灌胃;氟西汀组3 mg/kg灌胃;正常对照组和模型组每天给予同体积蒸馏水灌胃。采用强迫游泳、悬尾及高架十字迷宫实验法观察小鼠的行为学变化。结果与正常对照组比较,模型组的悬尾不动时间及强迫游泳不动时间均明显延长(P<0.01)。与正常对照组比较,小檗碱组强迫游泳不动时间缩短(P<0.05)。与模型组比较,小檗碱组的悬尾不动时间及强迫游泳不动时间均缩短(P<0.05);氟西汀组的悬尾不动时间及强迫游泳不动时间均明显缩短(P<0.01)。与正常对照组比较,模型组开臂区进入次数明显减少(P<0.05);与模型组比较,氟西汀组开臂区进入次数明显增多(P<0.01)。与正常对照组比较,模型组开臂区滞留时间明显缩短(P<0.05);与模型组比较,小檗碱组开臂区滞留时间明显延长(P<0.01),氟西汀组开臂区滞留时间明显延长(P<0.01)。结论小檗碱具有抗抑郁作用。Objective To investigate the effect of berberine on the behavioral changes in a mouse model of reserpine-induced depression.Methods A total of 40 experimental animals were randomly divided into the normal control group,the model group,the berberine group and the fluoxetine group,with 10 mice in each group.The mice in the model group,the berberine group and the fluoxetine group were intraperitoneally injected with 200 mg/kg reserpine(prepared in normal saline)every day,the normal control group was intraperitoneally injected with the same amount of normal saline every day,and the injection in each group lasted for 4 weeks.After 4 weeks of modeling,the drug was administered by gavage once a day for 21 days.The berberine group was given 190 mg/kg by gavage;the fluoxetine group was given 3 mg/kg by gavage;the normal control group and the model group were given the same volume of distilled water by gavage every day.The behavioral changes of mice were observed by forced swim test(FST),tail suspension test(TST)and elevated plus-maze test.Results Compared with the normal control group,the immobility time in TST and immobility time in FST in the model group were significantly longer(P<0.01).Compared with the normal control group,the im mobility time of forced swimming in the berberine group was shortened(P<0.05).Compared with the model group,the immobility time in TST and immobility time in FST in the berberine group were shorter(P<0.05);the immobility time in TST and immobility time in FST in the fluoxetine group were significantly shorter(P<0.01).Compared with the normal control group,the number of entries into the open arms in the model group was significantly reduced(P<0.05).Compared with the model group,the number of entries into the open arms in the fluoxetine group increased significantly(P<0.01).Compared with the normal control group,the time spent in the open arms in the model group was significantly shorter(P<0.05).Compared with the model group,the time spent in the open arms in the berberine group was significan
