摘 要:目的通过全氟丁基磺酸钾(PFBSK)急性暴露建立早期胚胎损伤模型,探讨PFBSK是否具有胚胎毒性及可能损伤的机制。方法本实验动物选取雄性10~12周龄和雌性6~8周龄的ICR小鼠,利用体外受精技术获取受精卵,并在含有PFBSK 0 mol/L(对照组)、10^(-3)mol/L、10^(-4)mol/L、10^(-5)mol/L及10^(-6)mol/L的培养基中培养受精卵,使其经历受精卵时期(1-细胞时期)、2-细胞时期、≥4-细胞时期、桑椹胚时期与囊胚时期,观察PFBSK是否对各个时期的胚胎发育率造成影响;并利用活性氧检测试剂盒和凋亡检测试剂盒分析是否存在由PFBSK引起的氧化应激损伤及细胞凋亡。结果与对照组相比,PFBSK 10^(-4)mol/L组、PFBSK 10^(-5)mol/L组和PFBSK 10^(-6)mol/L组的2-细胞率、≥4-细胞率、桑椹胚率及囊胚率均无显著性差异(P>0.05),并且胚胎发育形态良好,未见异常;而PFBSK 10^(-3)mol/L组的囊胚率与对照组相比显著下降(P<0.05),且部分胚胎停滞于桑椹胚,延长发育时期也无法使其生长至囊胚。PFBSK 10^(-3)mol/L组囊胚细胞活性氧水平显著高于对照组(P<0.05),但PFBSK 10^(-3)mol/L组的囊胚细胞凋亡平均荧光强度与对照组相比无显著性差异(P>0.05)。结论PFBSK可能存在潜在的胚胎毒性,会造成早期胚胎氧化应激损伤,但并非会走向主动凋亡的结局。Objective:To establish an early embryo injury model by acute exposure to perfluorobutane sulfonic acid potassium salt(PFBSK),and explore whether PFBSK has embryonic toxicity and its possible mechanism.Methods:In this experiment,ICR mice aged 10 to 12 weeks in males and 6 to 8 weeks in females were selected to obtain zygotes by in vitro fertilization.The zygotes were cultured in the medium containing PFBSK 0 mol/L(control group),10^(-3) mol/L,10^(-4) mol/L,10^(-5) mol/L and 10^(-6) mol/L,and the zygotes underwent the zygote stage,2-cell stage,≥4-cell stage,morula stage and blastocyst stage,to observe whether PFBSK influenced embryo development rate in each period.The reactive oxygen species detection kit and apoptosis detection kit were used to analyze whether there were oxidative stress injury and apoptosis caused by PFBSK.Results:Compared with the control group,there was no significant difference in 2-cell rate,≥4-cell rate,morula rate and blastocyst rate among PFBSK at 10^(-4) mol/L,10^(-4) mol/L and 10^(-4) mol/L(P>0.05),and the embryo development morphology was good,no abnormality is found.However,the blastocyst rate decreased at 10^(-3) mol/L(P<0.05),and some embryos stagnated in morula,and prolonging the development period could not make them grow to blastocyst.The level of reactive oxygen species in blastocyst cells in PFBSK 10^(-3) mol/L group was significantly higher than that in the control group(P<0.05),but the average fluorescence intensity of blastocyst cell apoptosis in PFBSK 10^(-3) mol/L group was not significantly different from that in the control group(P>0.05).Conclusions:PFBSK may be potentially toxic to embryos and may cause oxidative stress injury in early embryos,but it does not lead to active apoptosis.
