摘  要:为探究冷驯化条件下,胰岛素和甲状腺激素信号通路在中缅树鼩褐色脂肪组织(brown adipose tissue,BAT)中的调节机制,利用Illumina novaseq 6000平台对对照组和冷驯化组中缅树鼩BAT进行转录组测序,通过生物信息学分析筛选出与胰岛素和甲状腺激素相关的信号通路及候选基因。结果显示:冷驯化组较对照组,共2879个基因表达存在显著差异,其中1181个上调,1698个下调;GO分析结果显示细胞组分、分子功能、生物学过程分别显著富集到24、28和55条term,差异表达基因主要富集在生物学过程;KEGG分析差异表达基因显著富集到20条pathways,其中筛选到与胰岛素和甲状腺激素信号通路相关的候选基因有PDPK1、AKT2、ACACA、SLC2A4和THRB等。结果表明,中缅树鼩可通过调节BAT中胰岛素和甲状腺激素信号通路中基因表达来应对寒冷环境。In order to investigate the regulation mechanism of insulin and thyroid hormone signaling pathways in brown adipose tissue(BAT)in Tupaia belangeri under cold acclimation,in the present study,transcriptome sequencing of BAT in T.belangeri between control and cold acclimation group were carried out by Illumina novaseq 6000 platform,respectively.Then the signal pathways and candidate genes related to insulin and thyroid hormone were screened out by bioinformatics analysis.The results showed that there were significant differences in the expression of 2879 genes in cold acclimation group compared with that of control group,1181 were up-regulated and 1698 were down-regulated in cold acclimation group.The results of GO analysis showed that there were 24,28 and 55 terms were significantly enriched in cellular components,molecular functions and biological processes,respectively,and differentially expressed genes were mainly enriched in biological processes.KEGG analysis showed that the differentially expressed genes were significantly enriched in 20 pathways,and candidate genes related to insulin and thyroid hormone signaling pathways were screened out,including PDPK1,AKT2,ACACA,SLC2A4,and THRB,etc.All of the above results indicated that the T.belangeri could adapt to the cold environment by regulating genes expression in insulin and thyroid hormone pathways in BAT.
