摘 要:为了探究抗病毒蛋白Viperin对细胞自噬过程的影响以及该生物功能与物种间正向选择的相关性,利用分子生物学方法在人肾脏上皮细胞HEK293及肺癌细胞A549中过表达灵长类动物的Viperin,通过免疫荧光、Western blotting等方法检测Viperin对自噬相关蛋白表达的影响.结果表明:人类Viperin在HEK293细胞中的过表达诱导了胞内LC3由Ⅰ型向Ⅱ型的转化,且mCherry-GFP-LC3介导的胞内自噬体的形成量增加.自噬过程中的核心蛋白Beclin 1表达量明显增加,而敲低Beclin 1能减弱Viperin对自噬的诱导作用.同人类的Viperin相比,多种非人类灵长类动物的Viperin诱导LC3脂化的效果更强,Beclin 1的增加量更多,这表明抗病毒蛋白Viperin的生物学功能可能与灵长类动物进化中的正向选择过程相关.In order to explore the effect of the antiviral protein Viperin on the autophagic process and the correlation between its biological function and the positive selection among species,molecular biology methods were used to overexpress Viperin of different Primates in HEK293 and A549 cells.The effect of Viperin on the expression of autophagy-related proteins was detected by immunofluorescence,Western blotting and so on.The results showed that the overexpression of human Viperin in HEK293 cells induced the lipidation of intracellular LC3 from typeⅠto typeⅡ,and the formation of intracellular autophagosomes mediated by mCherry-GFP-LC3 increased.During autophagy,the expression of the autophagic core protein Beclin 1 was significantly up-regulated;Knockdown of Beclin 1 attenuated the induction of Viperin.Compared with Viperin of human,Viperin of non-human Primates has a stronger effect on inducing LC3 lipidation and the expression of Beclin 1.These results also suggest that the biological function of antiviral protein Viperin may be related to the positive selection in evolution of Primates.
