摘 要:目的:探讨娘母良对AD的治疗作用机制,并初步探讨娘母良在AD模型中对焦亡相关因子的调节作用。方法:将清洁级野生型C57B1/6J-Sv129雌性小鼠分为对照组(n=12),清洁级半合子3xTG雌性小鼠随机分为3组(n=12):模型组,阳性对照组,娘母良组。模型建立后通过Morris水迷宫实验评估动物的空间学习和记忆能力,采用酶联免疫吸附法(ELISA)检测小鼠海马区组织中Aβ_(40) 和Aβ_(42) 的浓度,通过TUNEL染色来验证各组小鼠模型神经细胞的凋亡,通过western blot检测小鼠海马区Tau蛋白异常磷酸化,并检测了焦亡因子caspase1(p20)以及GSDMD-N的相对蛋白表达。结果:①娘母良组与阳性对照组小鼠的逃避潜伏期,且随学习天数的增加而明显降低,到达隐藏平台的轨迹明显简化,且总路程以及总时间较模型组有明显下降;②娘母良组与阳性对照组小鼠的神经细胞的凋亡有明显的改善;③娘母良组与阳性对照组中小鼠的海马组织Aβ_(40) 和Aβ_(42) 浓度均明显下降;④娘母良组与阳性对照小鼠的海马组织p-Tau/Tau均明显下降;⑤娘母良组与阳性对照组小鼠的海马组织caspase1(p20)以及GSDMD-N的相对蛋白均明显下降。结论:娘母良可通过减少神经细胞凋亡,改善Aβ_(40) 、Aβ_(42) 沉积以及Tau蛋白异常磷酸化来改善AD模型鼠的空间学习和记忆能力,并且可能抑制AD模型鼠海马组织中神经细胞焦亡的发生。Objective To investigate the mechanism of therapeutic effects of Polygala crotalarioides on AD and to preliminarily explore the modulatory effects of Polygala crotalarioides on pyroptosis factors in an AD model.Methods Clean-grade wild-type C57B1/6J-Sv129 female mice were set as control group(n=12)and clean-grade 3xTG female mice were randomly divided into three groups(n=12):model group,positive control group and Polygala crotalarioides group.After model establishment,the spatial learning and memory abilities of the animals were assessed by Morris water maze experiment.The concentrations of Aβ_(40) and Aβ_(42) in the tissues of the hippocampal region of the mice were measured by enzyme-linked immunosorbent assay(ELISA),and the apoptosis of neuronal cells in each group was verified by TUNEL staining.The abnormal phosphorylation of Tau protein in the hippocampal region was detected by western blot,and the relative protein expression of the pyroptosis factors,cleaved caspase1(p20)and GSDMD-N,was detected.Results①The escape latency of mice in the Polygala crotalarioides group and the positive control group decreased significantly with the increase of learning days,the trajectory to the hidden platform was significantly simplified,and the total distance as well as the total time decreased significantly compared with the model group;②The apoptosis of neuronal cells in the Polygala crotalarioides group and the positive control group exhibited significant amelioration;③The concentration of Aβ_(40) and Aβ_(42) in the hippocampal tissue of mice in the Polygala crotalarioides group and the positive control group decreased significantly;④The p-Tau/Tau in hippocampal tissues of mice in the Polygala crotalarioides and positive control groups decreased significantly;⑤The relative protein of cleaved caspase1(p20)and GSDMD-N in hippocampal tissues of mice in the Polygala crotalarioides group and positive control groups decreased significantly.Conclusion Polygala crotalarioides group can meliorate the spatial learnin
