摘  要:目的:建立刷格痛风膏的质量标准。方法:采用TLC法鉴别刷格痛风膏中的黄连、大黄、黄柏;采用HPLC法测定盐酸小檗碱的含量;色谱柱为Inertsil ODS-3色谱柱(4.6 mm×250 mm Column,5μm);以乙腈-0.1%磷酸水溶液(28∶72)为流动相进行等度洗脱,流速1 mL/min,进样量2μL,检测波长265 nm,柱温25℃。结果:薄层色谱鉴别斑点清晰,阴性无干扰;盐酸小檗碱在0.004~0.1μg范围内呈现良好的线性关系(r=0.9998),平均回收率为99.45%,RSD为1.70%(n=6)。结论:该方法具有可行性,方法准确可靠,操作简单,重复性好,可作为刷格痛风膏的质量控制标准。Objective To establish the quality standard of Shuage Tongfeng Gao.Methods Rhizoma Coptidis、Rhei Radix et Rhizoma and cortex phellodendri were identified by TLC method.The content of Berberine Hydrochloride was determined by HPLC.The chromatographic separation was performed on a Inertsil ODS-3 Column(4.6×250 mm,5μm).The mobile phase consisted of acetonitrile-0.1%phosphoric acid(28∶72)with gradient elution at a flow rate of 1.0 mL·min^(-1),and the injection volume was 2μL.The detection wavelength was 265 nm.The Column temperature was at 25℃.Results The spots in TLC were clear without any interferences.Berberine Hydrochloride showed a good linear relationship within the range of 0.004~0.1μg(r=0.9998),the average recovery rate was 99.45%,and the RSD was 1.7%(n=6).Conclusion The established qualitative and quantitative methods are simple,reliable and accurate,which can be used for the quality control of Shuage Tongfeng Gao.
