摘 要:目的探究乳化异氟醚对小鼠睾丸间质细胞生物学功能的影响及其分子机制。方法将小鼠睾丸间质细胞TM3分为对照组、低剂量乳化异氟醚组、中剂量乳化异氟醚组和高剂量乳化异氟醚组。对照组加入30%脂肪乳,低、中和高剂量乳化异氟醚组分别采用终浓度0.5、1和2 mmol/L乳化异氟醚进行处理。采用MTT法检测细胞活力,流式细胞术检测细胞凋亡率,Western blot检测Bax/Bcl-2、p-Akt/Akt和p-PI3K/PI3K的表达,酶联免疫吸附实验(enzyme-linked immunosorbent assay,ELISA)检测细胞睾酮分泌情况,比色法检测超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和丙二醛(MDA)水平。结果与对照组相比,低、中和高剂量乳化异氟醚组小鼠睾丸间质细胞的细胞活力、睾酮分泌量、SOD与CAT水平均明显降低(P<0.05),细胞凋亡率,Bax/Bcl-2、p-Akt/Akt和p-PI3K/PI3K表达水平,MDA水平均明显增加(P<0.05)。PI3K抑制剂LY294002可逆转乳化异氟醚对小鼠睾丸间质细胞的细胞活力,凋亡率,Bax/Bcl-2、p-Akt/Akt、p-PI3K/PI3K表达水平,睾酮分泌量和SOD、CAT、MDA水平的影响(P<0.05)。结论乳化异氟醚通过激活PI3K/AKT信号通路引发小鼠睾丸间质细胞损伤。Objective To investigate the effect of emulsified isoflurane on biological function of mouse testicular stromal cells and its molecular mechanism.Methods Mouse testicular stromal cell TM3 was divided into control group,low dose emulsified isoflurane group,medium dose emulsified isoflurane group and high dose emulsified isoflurane group.The control group was treated with 30%fat emulsion.The low-dose emulsified isoflurane group,medium-dose emulsified isoflurane group and the high-dose emulsified isoflurane group were treated with emulsified isoflurane with a final concentration of 0.5 mmol/L,1 mmol/L and 2 mmol/L respectively.MTT assay was used to detect cell viability,flow cytometry was used to detect apoptosis rate,Western blot was used to detect the expression of Bax/Bcl-2,p-Akt/Akt and p-PI3 K/PI3 K,enzyme-linked immunosorbent assay was used to detect testosterone secretion,colorimetry was used to detect the level of SOD,CAT,MDA.Results Compared with control group,cell viability,testosterone secretion,SOD and CAT levels of testicular interstitial cells in low,medium and high dose emulsified isoflurane groups were significantly decreased(P<0.05),while the apoptosis rate,Bax/Bcl-2,p-Akt/Akt,P-PI3 K/PI3 K expression level and MDA level were significantly increased(P<0.05).PI3 K inhibitor LY294002 could reverse the effects of emulsified isoflurane on cell viability,apoptosis rate,expression levels of Bax/Bcl-2,P-Akt/Akt,P-PI3 K/PI3 K,testosterone secretion,SOD,CAT and MDA of mouse testicular interstitial cells(P<0.05).Conclusion Emulsified isoflurane induced damage to mouse testicular stromal cells by activating PI3 K/AKT signaling pathway.
