摘 要:目的探讨PM_(2.5)诱导的心肌损伤及硒的保护作用。方法健康清洁级雄性SD大鼠32只,随机分为4组,对照组、PM_(2.5)组、硒组及硒+PM_(2.5)组。分别灌胃给予生理盐水1 ml,或亚硒酸钠溶液35μg/kg·bw后,利用定量雾化器给予1.5 ml生理盐水或140.16μg/ml PM_(2.5)悬液,连续30 d,禁食过夜后,麻醉取血及心脏组织,利用相关试剂盒检测相应指标。结果大鼠暴露PM_(2.5)后,体重增加值较对照组少,硒预处理有所改善;大鼠暴露PM_(2.5),其心肌组织中谷光甘肽过氧化物酶(GSH-Px)及总超氧化物歧化酶(T-SOD)水平较对照组明显降低(P<0.05),丙二醛(MDA)水平较对照组明显增加(P<0.05),硒预处理后,GSH-Px水平较PM_(2.5)组明显升高(P<0.05),MDA水平较PM_(2.5)组明显降低,差异有统计学意义(P<0.05);PM_(2.5)可使大鼠心肌组织中一氧化氮(NO)含量及一氧化氮合酶(NOS)活力水平明显升高(P<0.05),硒预处理可明显降低NO含量及NOS活力水平(P<0.05);PM_(2.5)染毒组大鼠血清中乳酸脱氢酶(LDH)、天冬氨酸转氨酶(AST)、肌酸激酶(CK)及心肌肌钙蛋白T(cTnT)水平明显升高(P<0.05),硒预处理可明显降低这些指标的水平,差异有统计学意义(P<0.05)。结论大鼠暴露一定剂量PM_(2.5)可导致心脏受损,一定剂量硒的干预可在一定程度上起到缓解作用。Objective To investigate the myocardial injury induced by PM_(2.5) and the protective effect of selenium.MethodsA total of 32 SD male rats were randomly divided into four groups:control group,PM_(2.5) group,selenium intervention group and selenium+PM_(2.5) group.1 ml normal saline or 35μg/kg·bw sodium selenite solution was given to rats by gavage,then 1.5 ml normal saline or 140.16μg/kg·bw PM_(2.5) suspension was given via tracheal instillation by using of lung quantitative nebulizer for 30 days.After fasting overnight,blood and cardiac tissue were collected under anesthesia.Some related indexes were detected by relevant kits.Results The body weight gain of rats exposed to PM_(2.5) was less than that of control group,and selenium intervention improved this condition;The levels of GSH-Px and T-SOD in myocardial tissue of PM_(2.5) group were significantly lower than those in control group(P<0.05),and MDA level was significantly increased compared with the control(P<0.05).After selenium intervention,level of GSH-Px was significantly higher than that of PM_(2.5)group(P<0.05),and MDA level was significantly lower than that of PM_(2.5)group(P<0.05);PM_(2.5)could significantly increase the content of NO and the activity of NOS in myocardial tissue(P<0.05),while selenium intervention could significantly reduce the level of NO and NOS activity(P<0.05);The levels of LDH,AST,CK and cTnT in serum of PM_(2.5)group were significantly increased(P<0.05),selenium intervention could significantly dcrease the levels of LDH,CK and cTnT(P<0.05).Conclusion Exposure to a certain dose of PM_(2.5)can cause heart damage,and the intervention of certain dose of selenium can alleviate the damage to a certain extent.
