摘 要:目的:建立食物中毒样品中大肠埃希氏菌(Escherichia coli)的检测方法,并对分离的大肠埃希氏菌进行生化鉴定和药敏分析。方法:根据E. coli特异性基因设计引物和探针序列,建立荧光定量PCR检测体系,并使用VITEK系统对分离的E. coli进行生化鉴定和药敏分析。结果:成功建立了荧光定量PCR检测E. coli的方法,方法检测灵敏度高(LOD为10 CFU·mL^(-1))、特异性强,应用该方法对10份食物中毒样品中的E. coli进行检测,发现4份阳性样本,纯培养后获取4株E. coli,进一步的VITEK生化鉴定结果表明这4株分离株为E. coli,药敏分析结果显示4株E. coli分离株均表现为多重耐药株。结论:本研究建立的荧光定量PCR能够灵敏、准确地检测食物中毒样品中的E. coli,且通过生化鉴定和药敏分析掌握食物中毒样品中E. coli的生化特征,为预防和控制E. coli引发的食物中毒提供了科学依据。Objective: To establish a detection method for Escherichia coli in food poisoning samples, and to carry out biochemical identification and drug susceptibility analysis of the isolated E. coli. Method: The primers and probe sequences were designed according to the specific genes of E. coli, and a fluorescence quantitative PCR detection system was established. The VITEK system was used to carry out biochemical identification and drug susceptibility analysis of the isolated E. coli. Result: A fluorescent quantitative PCR method for the detection of E. coli was successfully established, with high detection sensitivity(LOD of 10 CFU·mL^(-1)) and strong specificity. The method was used to detect E. coli in 10 food poisoning samples, and four were found. Four positive samples were obtained after pure culture. Further VITEK biochemical identification results showed that these four isolates were E. coli, and the drug susceptibility analysis showed that the four E.coli isolates were all multi-drug resistant strains. Conclusion: The fluorescence quantitative PCR established in this study can sensitively and accurately detect E. coli in food poisoning samples, and the biochemical characteristics of E. coli in food poisoning samples can be mastered through biochemical identification and drug susceptibility analysis, in order to prevent and control E. coli. Food poisoning caused by coli provides a scientific basis.
