摘 要:目的探讨在M-Sol中添加海藻糖对体外4℃保存血小板的影响。方法随机选取7袋ABO同型浓缩血小板汇集后,根据保存温度、保存介质和海藻糖浓度的不同均分成7组,分别为22℃+PRP组、PRP组、M-Sol+PRP组、M-Sol+PRP+1 g/L海藻糖组、M-Sol+PRP+5 g/L海藻糖组、M-Sol+PRP+15 g/L海藻糖组和M-Sol+PRP+25 g/L海藻糖组。PRP组及M-Sol保存各组均置于4℃保存,分别于保存d1、3、5、7取样检测Plt、PDW、MPV、最大聚集度、CD62p阳性率和GLU及LAC浓度,并计算7 d内GLU及LAC浓度变化量。结果随着保存时间的延长,各组Plt均下降,且同时间点各组间比较无统计学差异(P>0.05),各组PDW、MPV均有升高,保存到d7时,以M-Sol+PRP+5 g/L海藻糖组的PDW 10.22±0.43(fL)、MPV 8.74±0.40(fL)最低,与22℃+PRP组比较有统计学差异(P<0.05)。7 d内的GLU及LAC浓度变化量均以M-Sol+PRP+5 g/L海藻糖组最低,其7 d内的GLU浓度变化量与22℃+PRP组比较有统计学差异(P<0.05),7 d内的LAC浓度变化量与PRP组比较无统计学差异(P>0.05)。保存期间,各组最大聚集度逐渐下降,保存d7时,除PRP组外,M-Sol+PRP+5 g/L海藻糖组最大聚集度最高(27.29±6.62),差异与22℃+PRP组比较有统计学意义(P<0.05)。保存d7时,M-Sol+PRP+5 g/L海藻糖组的CD62p阳性率最低(15.46±2.46),与其他组比较均有统计学差异(P<0.05)。结论M-Sol中添加适宜浓度的海藻糖可抑制4℃血小板的活化,降低血小板的储存损伤。Objective To investigate the effect of trehalose added to M-Sol on platelet preservation at 4℃in vitro.Methods Seven bags of ABO homotype concentrated platelets were randomly selected and divided into 7 groups according to storage temperature,preservation medium and trehalose concentration:22℃+PRP group,PRP group,M-Sol+PRP group,M-Sol+PRP+1 g/L trehalose group,M-Sol+PRP+5 g/L trehalose group,M-Sol+PRP+15 g/L trehalose group and M-Sol+PRP+25 g/L trehalose group.PRP group and M-Sol preservation groups were stored at 4℃.Plt,PDW,MPV,maximum aggregation,CD62 p positive rate,GLU and LAC concentration were detected on the 1 st,3 rd,5 th and 7 th day after preservation,and the changes of GLU and LAC concentration within 7 days were calculated.Results With the extension of preservation time,Plt decreased in all groups,and there was no significant difference among groups at the same time(P>0.05).PDW and MPV increased in all groups.When preserved to the 7 th day,the PDW 10.22±0.43(fL)and MPV 8.74±0.40(fL)of M-Sol+PRP+5 g/L trehalose group were the lowest,which was significantly different from that of 22℃+PRP group(P<0.05).The changes of GLU and LAC concentration within 7 days were the lowest in M-Sol+PRP+5 g/L trehalose group,and the change of GLU concentration within 7 days was significantly different from that of 22℃+PRP group(P<0.05),but not significantly different from that of PRP group(P>0.05).During the preservation period,the maximum aggregation degree of each group decreased gradually.Except for the PRP group,the maximum aggregation degree of the M-Sol+PRP+5 g/L trehalose group was the highest(27.29±6.62),which was significantly higher than that of the 22℃+PRP group on the 7 th day after preservation(P<0.05).On the 7 th day after preservation,the positive rate of CD62 p in M-Sol+PRP+5 g/L trehalose group was the lowest(15.46±2.46),and there was significantly different compared with other groups(P<0.05).Conclusion Adding appropriate concentration of trehalose to M-Sol can inhibit platelet activation a
