摘 要:目的:研发一种具有靶向肿瘤功能的中药纳米脂质体并考察其体外抗肿瘤效应。方法:采用薄膜分散法制备靶向EpCAM的适配体(SYL3C)功能化绞股蓝皂苷(Gyp)脂质体SYL3C-Gyp@Lipo。考察SYL3C-Gyp@Lipo的表征特性,采取流式细胞仪检测其靶向性,MTT法评价其对人肝癌细胞HepG2的细胞毒性。结果:成功制备一种能够靶向肝癌的绞股蓝皂苷脂质体SYL3C-Gyp@Lipo,其分散性良好,呈圆球形或椭圆球形,能够减缓药物的释放,在体外能特异性杀伤肝癌HepG2细胞。结论:SYL3C-Gyp@Lipo能靶向杀伤高表达EpCAM的肝癌细胞,达到肿瘤靶向治疗效果,为制备新型的中药纳米载药制剂提供了一种潜在策略。Objective:To develop a traditional Chinese medicine nano-liposome with tumor targeting function and to investigate its anti-tumor effect in vitro.Methods:EpCAM-targeting aptamer(SYL3C) functionalized gynostemma pentaphyllum(Gyp) liposome SYL3C-Gyp@Lipo was prepared by thin film dispersion method.The characterization of SYL3C-Gyp@Lipo was investigated,its targeting was detected by flow cytometry,and its cytotoxicity to human hepatocellular carcinoma cell line HepG2 was evaluated by MTT method.Results:A kind of gynostemma pentaphyllum liposome SYL3C-Gyp@Lipo was successfully prepared,which can target liver cancer.It had good dispersion with spherical or ellipsoidal shape,could slow down the drug release,and specifically kill hepatoma HepG2 cells in vitro.Conclusion:SYL3C-Gyp@Lipo can target hepatoma cells with high expression of EpCAM and achieve the effect of tumor targeted therapy,which provides a potential strategy for the preparation of new nano-drug-loaded preparations of traditional Chinese medicine.
