轮叶党参总皂苷对糖尿病大鼠心肌保护作用的研究

摘  要:目的:探讨轮叶党参总皂苷(CLTS)对糖尿病(DM)大鼠心肌的保护作用。方法:构建2型糖尿病(T2DM)大鼠模型。将大鼠随机分为正常对照组(正常大鼠)、模型组和轮叶党参组。对照组及模型组大鼠常规用生理盐水灌胃,轮叶党参组大鼠用CLTS水溶液灌胃,共灌胃8周。末次灌胃后将三组大鼠断头处死,分离心脏,并采用酶联免疫吸附法(ELISA)检测其心肌组织匀浆上清液中肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)、丙二醛(MDA)、可溶性细胞间粘附分子-1(sLCAM-1)及可溶性血管间粘附分子-1(sVCAM-1)的表达水平。结果:正常对照组大鼠和轮叶党参组大鼠心肌组织中SOD的表达水平均高于模型组大鼠,MDA和TNF-α的表达水平均低于模型组大鼠,差异有统计学意义(P<0.01或P<0.05)。轮叶党参组大鼠心肌组织中SOD的表达水平低于正常对照组大鼠,MDA和TNF-α的表达水平均高于正常对照组大鼠,差异有统计学意义(P<0.01或P<0.05)。正常对照组大鼠和轮叶党参组大鼠心肌组织中sLCAM-1、sVCAM-1的表达水平均低于模型组大鼠,差异有统计学意义(P<0.05)。轮叶党参组大鼠心肌组织中sLCAM-1、sVCAM-1的表达水平均高于正常对照组大鼠,差异有统计学意义(P<0.05)。结论:CLTS可显著改善DM大鼠心肌组织中TNF-α、SOD、MDA、sLCAM-1和sVCAM-1的表达水平,保护其心肌组织,降低其心血管疾病的发病风险。Objective:To investigate the protective effect of total saponins(CLTS)of Codonopsis folium on myocardial of diabetic rats.Methods:The rat model of type 2 diabetes mellitus(T2DM)was established.The rats were randomly divided into normal control group(normal rats),model group and Codonopsis rotundus group.Rats in control group and model group were intragastric with normal saline,and rats in Codonax rotundus group were intragastric with CLTS solution for 8 weeks.After the last intragastric administration,the rats in the three groups were decapitated and their hearts were separated.The expression levels of tumor necrosis factor-α(TNF-α),superoxide dismutase(SOD),malondialdehyde(MDA),soluble intercellular adhesion molecule-1(SLCAM-1)and soluble intervascular adhesion molecule-1(SVCAM-1)in the supernatant of myocardial tissue homogenate were determined by enzyme-linked immunosorbent assay(ELISA).Results:The expression level of SOD in myocardium of normal control group and COdonopsis rofolia group was higher than that of model group,while the expression leve l of MDA and TNF-αwas lower than that of model group,the differences were statistically significant(P<0.01 or P<0.05).The expression level of SOD and MDA and TNF-αin myocardium of L.l.group was lower than that of normal control group,and the differences were statistically significant(P<0.01 or P<0.05).The expression levels of SLCAM-1 and SVCAM-1 in myocardium of normal control group and COdonopsis rotundus group were lower than that of model group,with statistical significance(P<0.05).The expression levels of SLCAM-1 and SVCAM-1 in myocard ium of S.lunyatidis group were higher than those of normal control group,with statistical significance(P<0.05).Conclusion:CLTS can significantly improve the expression levels of TNF-α,SOD,MDA,SLCAM-1 and SVCAM-1 in myocardial tissue of DM rats,protect the myocardial tissue,and reduce the risk of cardiovascular disease.
