摘  要:为了研究鹅肌肉生长抑制素(myostatin, MSTN)基因的多态性及其与体重的关系,试验采用DNA直接测序技术,检测莱茵鹅和籽鹅MSTN基因的单核苷酸多态性(SNPs)位点,应用PopGen32和PIC-CALC软件计算遗传多态参数及进行卡方适合性检验,利用SPSS 17.0软件的GLM(General Linear Model)模型分析基因型对体重的遗传效应。结果表明:在2个鹅品种MSTN基因中共检测到2处突变,分别是位于启动子区域的c.-490T>G(P1位点),产生3种基因型,即TT、TG和GG;位于外显子1区域的c.84G>C突变(E1位点),产生3种基因型,即GG、GC和CC。莱茵鹅在P1和E1位点极显著偏离哈代-温伯格平衡(P<0.01);籽鹅在P1位点处于哈代-温伯格平衡状态(P>0.05),在E1位点极显著偏离哈代-温伯格平衡(P<0.01)。对于MSTN基因P1位点,除了3,4周龄外,GG基因型莱茵鹅个体体重显著或极显著高于TT基因型个体(P<0.01或P<0.05)。对于MSTN基因E1位点,初生、1,2,10~12周龄CC基因型莱茵鹅个体体重显著或极显著高于GG基因型个体(P<0.05或P<0.01);初生、1周龄CC基因型籽鹅个体体重极显著高于GG基因型个体(P<0.01)。说明MSTN基因的SNPs位点有可能成为莱茵鹅和籽鹅体重选育的分子标记。In order to study the polymorphism of myostatin(MSTN) gene in geese and its relationship with weight, single nucleotide polymorphisms(SNPs) of MSTN gene in Rhine geese and Zi geese were detected by DNA direct sequencing. Genetic polymorphism parameters were calculated by PopGen32 and PIC-Calc software and chi-square test was calculated. The genetic effects of genotypes on weight were analyzed by General Linear Model(GLM) of software SPSS 17. 0.The results showed that two mutations were detected in the MSTN gene of the two goose breeds, which were c.-490 T>G located in the promoter region, producing 3 genotypes, namely TT、TG and GG. The c.84 G>C mutation(E1 locus) located in exon 1 region produced 3 genotypes, namely GG、GC and CC. Rhine geese deviated significantly from hardy Weinberg equilibrium at P1 and E1 locus(P< 0.01). The Zi geese were in Hardy Weinberg equilibrium at P1 locus(P>0.05), and deviated significantly from hardy Weinberg equilibrium at E1 locus(P< 0.01). For the P1 locus of MSTN gene, the body weight of Rhine geese with GG genotype was significantly or very significantly higher than that with TT genotype except for body weight at 3 and 4 weeks(P<0.01 or P<0.05). For E1 locus of MSTN gene, the body weight of Rhine geese with CC genotype was significantly or very significantly higher than that with GG genotype at birth, 1, 2, and 10-12 weeks old(P<0.01 or P<0.05). The body weight of Zi geese with CC genotype was significantly or very significantly higher than that with GG genotype at birth and 1 week. The results indicated that the SNPs locus of MSTN gene might be a molecular marker for weight selection in Rhine geese and Zi geese.
