摘 要:为了解吉林市周边猪场弓形虫的感染情况及其基因型分型,试验于2019年10月份—2020年9月份在吉林市周边9个养猪场采集326份血清样本,利用ELISA方法进行血清中弓形虫抗体检测,计算猪场弓形虫阳性率。利用动物试验进行阳性虫株的分离并进行细胞培养,利用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法在SAG3、GRA6和L358 3个位点进行分离虫株的基因型初步鉴定。结果表明:吉林市周边猪场血清学检测弓形虫平均阳性率为28.31%;分离得到1株虫株,命名为JLpig1株。临床分离株JLpig1在位点GRA6处表现为基因Ⅰ型和基因Ⅱ型;在位点SAG3处表现为基因Ⅰ型和基因Ⅱ型;在位点L358处表现为基因Ⅱ型和China1型(Toxo DB#9株)。初步判断吉林分离株JLpig1是强毒株或中毒株可能性较大,以基因Ⅱ型为主。说明吉林市周边猪场弓形虫病感染较为普遍,且感染率较高,应提高猪场人员对猪弓形虫病的认识,加强对猪场猪弓形虫病的防控。In order to understand the infection and genotyping of Toxoplasma gondii in surrounding pig farms in Jilin City, 326 serum samples were collected from 9 pig farms around Jilin City from October 2019 to September 2020. ELISA method was used to detect Toxoplasma gondii antibody in serum, and the positive rate of Toxoplasma gondii in pig farms was calculated. Animal testing was used to isolate positive strain and cell culture was carried out. The genotypes of the isolates were preliminarily identified at three sites of SAG3, GRA6 and L358 by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). The results showed that the average positive rate of serological detection of Toxoplasma gondii was 28.31% in pig farms around Jilin City;one strain was isolated and named as JLpig1 strain. The clinical isolate JLpig1 showed genotype Ⅰ and genotype Ⅱ at site GRA6;genotype Ⅰ and genotype Ⅱ at site SAG3;genotype Ⅱ and China1 at site L358(Toxo DB#9) type. It was preliminarily judged that the Jilin isolate JLpig1 was a virulent strain or a moderately virulent strain, which was mainly genotype Ⅱ. The results suggested that toxoplasmosis infection was common in pig farms around Jilin City, and the infection rate was high. It was necessary to improve the knowledge of pig farm personnel on pig toxoplasmosis, and to strengthen the prevention and control of toxoplasmosis in pig farms.
