摘  要:目的:探究不同剂量麦芽总生物碱(total barley maiya alkaloids,TBMA)对产后缺乳大鼠乳汁分泌的影响及机制。方法:产后24 h内SD大鼠随机分为对照组、模型组、胃复安组、7.5 g组(成人日服7.5 g生麦芽所含TBMA,下同)、15 g组、30 g组、60 g组,灌胃溴隐亭建立产后缺乳大鼠模型,各组灌胃相应药物,持续12 d。观察母鼠泌乳量、仔鼠12 d窝重净增量;用HE染色观察各母鼠乳腺组织病理变化;ELISA测定各组大鼠血清泌乳素(prolactin,PRL)、雌二醇(sstradiol2,E2)、孕酮(progesterone,P)水平;Western Blot检测母乳腺组织PRL、泌乳素受体(prolactin receptor,PRLR)、β-酪蛋白(β-casein)蛋白表达;免疫组化检测母鼠乳腺组织PRLR、非受体型酪氨酸蛋白激酶2(janus kinase 2,JAK2)、信号传导及转录激活蛋白5(signal transducer and activator of transcription 5,Stat5)蛋白表达。结果:15 g组母鼠泌乳量和仔鼠12 d窝重净增量显著升高(P<0.05、P<0.01),大鼠乳腺小叶明显扩张、乳汁充盈,血清PRL、E2、P水平均显著升高(P<0.05、P<0.01),乳腺组织中PRL、β-casein、PRLR蛋白表达明显升高(P<0.05),JAK2、Stat5蛋白表达升高;而7.5,30,60 g组均无上述作用。结论:15 g生麦芽所含TBMA能有效促进泌乳,其作用机制可能与由PRLR介导的JAK2/Stat5信号通路有关。OBJECTIVE To investigate the effects of different doses of total malt maiya alkaloids(TBMA)on the milk secretion of postpartum lactogenic rats and its mechanism.METHODS SD rats were randomly divided into control group,model group,metoclopramide group group 7.5 g(adult daily dose of total malt alkaloid contained in 7.5 g raw malt,the same below)group 15 g,group30 g,group60 g within24 h after delivery,bromocriptine methylate was given intragastrically(intragastrically)to establish postpartum hypogalactosis rat model,each group was given relevant drugs intragastrically for 12 days.Milk yield of female rats and net increase of litter weight of newborn rats at 12 days were observed.HE staining was used to observe the pathological changes of mammary gland tissues.Serum levels of prolactin(PRL),sstradiol2(E2),progesterone(P)were determined by ELISA.Western Blotting was used to detect the protein expression of PRL,Prolactin receptor(PRLR)andβ-casein in female breast tissue.PRLR,Janus kinase 2(JAK2),signal and activator of transcription 5(PRLR)in mammary tissues of female rats were detected by immunohistochemistry protein expression.RESULTS The net increase of lactation and litter weight at 12 days in 15 g group were significantly increased(P<0.05,P<0.01),the lobules of mammary gland were significantly dilated,milk was filled,serum PRL,E2,P levels were significantly increased(P<0.05,P<0.01),the protein expression of PRL,β-casein and PRLR in breast tissue were significantly increased(P<0.05),JAK2,Stat5 protein expression increased;However,7.5,30 and 60 g groups had no such effects.CONCLUSION Total malt alkaloids contained in 15 g raw malt can effectively promote lactation,and its mechanism may be related to JAK2/Stat5 signaling pathway mediated by PRLR.
