左归丸治疗骨质疏松的网络药理学分析

摘　　要：目的:运用网络药理学方法探讨左归丸治疗骨质疏松的潜在药效机制。方法:从《中华人民共和国药典》收集左归丸中各中药的归经信息,TCMSP、BATMAN-TCM数据库中查找中药活性成分和靶标,Drugbank、DisGeNET数据库中查找骨质疏松的潜在靶点,利用Cytoscape软件构建和分析中药-归经网络、靶标-疾病网络,利用DAVID对靶标-疾病网络中最大连通片子模块节点基因进行功能富集分析。结果:左归丸所含中药主要归肾、肝经,其潜在活性化合物73个、51个靶标与骨质疏松相关,靶标-疾病网络中3个关键子模块主要生物学功能涉及转化生长因子β(transforming growth factor-β,TGF-beta)、雌激素(Estrogen)、甲状腺激素(Thyroid hormone)、磷脂酰肌醇3激酶-蛋白激酶B(phosphatidylinositol 3-kinase and protein kinase B,PI3K-Akt)等16个信号通路,影响骨代谢平衡。结论:左归丸“补肾壮骨”法治疗骨质疏松,可能通过调控骨代谢平衡中成骨、破骨分化多个生物学过程发挥治疗作用。Objective:To explore the pharmacological mechanism of Zuogui Piu in the treatment of Osteoporosis based on network pharmacology.Methods:The meridian tropism of the herbs in Zuogui Piu was collected from the Pharmacopoeia of the People's Republic of China.The compounds and targets of each herb were collected from TCMSP or BATMAN-TCM databases,and the genes of osteoporosis were collected from Drugbank and DisGeNET.Cytoscape software was used to construct and analyze the herb-meridian network and target-disease network.DAVID was applied for functional enrichment analysis of the maximum connected chip genes in the targets-diseases networks.Results:The top meridian tropism of herbs in Zuogui Piu was kidney and liver meridian,Among Zuogui Piu,there are 73 putative targets,51 targets were co-expressed with osteoporosis.In the target-disease network,there are 3 key submodules,including 16 biological functions,such as TGF-β,Estrogen,Thyroid hormone,PI3K-Akt signaling pathways in bone metabolic balance.Conclusion:Zuogui Piu treats osteoporosis with the therapy of kidney-reinforcing and bone-strengthening,which maybe regulate osteogenesis and osteoclast differentiation in bone metabolic balance through several biological processes.
