雌孕激素联合对实验性大鼠口腔溃疡的治疗作用

摘　　要：口腔溃疡是常见的口腔黏膜疾病之一,内分泌因素被认为是诱发口腔溃疡的病因之一,本研究探讨雌激素联合孕激素对大鼠口腔溃疡的治疗作用。以Na OH刺激方法制备口腔溃疡大鼠模型,用雌二醇和黄体酮单独或联合局部干预10 d(2次/d),每天观察溃疡改变,绘制溃疡愈合曲线。10 d后取溃疡组织做HE和Masson染色检测病理和胶原成分变化。运用免疫组化和Real-Time PCR方法检测溃疡组织TGF-β1、Keratin 5的蛋白以及mRNA改变。结果显示:相较于模型组和单独给药干预组,雌激素联合孕激素可以促进口腔溃疡的愈合,增加溃疡组织胶原含量,增加TGF-β1、Keratin 5的mRNA和蛋白表达。本研究初步证实了雌激素联合孕激素局部处理可以促进大鼠口腔溃疡的愈合,该作用与增加溃疡组织胶原合成、促进上皮细胞增殖和角化有关,并提示内分泌改变可能是口腔溃疡的一个重要因素。Oral ulcer is one of the common oral mucosal diseases, and mouth ulcers are associated with menstrual period in female population. This study was to investigate the therapeutic effect of estrogen combined with progesterone on oral ulcer in rat model. The rat oral ulcer model was prepared by Na OH stimulation method, and local intervention with estradiol and progesterone alone or in combination for 10 days(twice a day). Observed ulcer changes daily and drew the ulcer healing curves. After 10 days, the ulcer tissues were taken for HE and Masson staining to detect pathological changes and collagen composition. Protein and mRNA changes of TGF-β1 and Keratin 5 in ulcer tissues were detected by immunohistochemistry and Real-time PCR method. The results showed that estrogen combined with progesterone could promote the healing of oral ulcer, increase the collagen content of ulcer tissue, and increase the mRNA and protein expression of TGF-β1 and Keratin 5, compared with the model group and single administration intervention group. This study initially confirmed that local treatment of estrogen combined with progesterone promote the healing of oral ulcer in rats, and this effect is related to the increase of collagen synthesis in ulcer tissue and the promotion of epithelial cell proliferation and maturation, suggesting that endocrine changes may be an important factor in oral ulcer.
