摘 要:目的:通过蒙医震脑术治疗脑震荡大鼠,观察对脑震荡大鼠脑组织神经递质的影响,从而阐述作用机制及原理。方法:将80只SPF等级的SD大鼠[雄性、体质量(250±20)g]随机分为10组:正常组、模型4 d组、模型7 d组、模型10 d组、西药4 d组、西药7 d组、西药10 d组、2次震脑术组、3次震脑术组、4次震脑术组。造模前后以及治疗后进行行为学测试,以评估认知功能、运动功能、焦虑和抑郁。在实验第4天、第7天、第10天分别取材,用Western Blot方法检测大鼠海马、皮质及下丘脑内5HT和Ach的含量。结果:Western Blot方法结果表明,大鼠脑震荡后神经递质含量会发生改变,蒙医震脑术可以调节大鼠脑组织神经递质,从而减少脑组织的损伤,达到治疗和减轻症状以及减少脑震荡后遗症的目的。结论:本次实验发现暗示了神经递质在脑震荡后的病理生理学后果,并提示蒙医震脑术是一种可行的治疗选择,以减轻这些影响及其有害后果。Objective: To observe the effects of brain tissue and immune function with concussion rats treated by brain vibration therapy, and to expounds the mechanism and principle of the treatment concussion by Mongolian medicine brain vibration therapy. Methods: The 80 SPF-grade SD rats(male, weighing 250 ±20g) were randomly divided into ten groups:control group, model 4 days, model 7 days, model 10 days, western medicine 4 days, western medicine 7 days, western medicine 10 days, 2 brain vibration therapy group, 3 brain vibration therapy group, 4 brain vibration therapy group, a total of10 groups. Behavioral tests were performed before and after modeling and after treatment to evaluate cognitive function, motor function, anxiety and depression. On the fourth, seventh and tenth day of the experiment, the contents of 5-HT and Ach in hippocampus, cortex and hypothalamus were detected by Western blot. Results: Western blot results showed that the content of neurotransmitters in rats after cerebral concussion would change. Mongolian medicine brain vibration therapy can regulate the neurotransmitters in rats’ brain tissue, so as to reduce the damage of brain tissue, and achieve the effect of treating and alleviating symptoms and reducing the sequelae of cerebral concussion. Conclusion: This experiment found and implied the pathophysiological consequences of neurotransmitters after cerebral concussion, and suggested that Mongolian medicine brain vibration therapy is a feasible treatment option to reduce these extremely harmful consequences.
