摘  要:目的:通过数据挖掘老年性肺炎中医药治疗方案,基于“中医传承计算平台V3.0”软件及网络药理学分析老年性肺炎方证规律及作用机制,为临床辨证用药和相关研究提供参考信息。方法:通过在中国知识资源总库、万方数据知识服务平台、维普网检索栏中以“老年性肺炎”“中医药”等为主题词进行搜索,选取中医参与治疗的案例进行汇总,采用中医传承计算平台分析治疗老年性肺炎处方的用药规律,并进一步探讨该病病机及演变规律。通过中药系统药理学数据库与分析平台(Traditional Chinese Medicine System Pharmacology Database and Analysis Platform,TCMSP)、中药分子机制研究生物信息学分析工具寻找与肺炎使用频次前15味中药相关的所有有效成分和作用靶点,并通过TTD、Drug Bank、Dis Ge NET、Gene Cards数据库筛选与肺炎相关的靶点,构建药物-成分-靶点-疾病网络,利用STRING平台构建药物-疾病靶点交互网络,使用Omic Share云平台对靶点蛋白进行基因本体论(GO)富集分析及京都基因与基因组百科全书(KEGG)通路富集分析,分子对接分析使用Autodock Vina。结果:从数据库中共检索到相关文献519篇,经过筛选得到81篇文献,涉及42首治疗老年性肺炎中医古方,其中麻杏石甘汤(13次)、千金苇茎汤(或苇茎汤)(10次)、补肺汤(9次)、清金化痰汤(8次)使用频次较多,共涉及自拟方25首及6种中成药,其中甘草、黄芩、杏仁、茯苓、桑白皮、桔梗、麦门冬、陈皮等使用频次较高,通过平台关联规则分析得到黄芩-甘草、杏仁-黄芩、黄芩-鱼腥草、桔梗-甘草、黄芩-桔梗、杏仁-甘草等常用药物组合。通过网络药理学得到329个活性成分,其中具有靶点的有效成分275个,共得到463个药物潜在靶点;4个疾病数据库共收集到1367个疾病靶点,药物有效成分靶点与疾病靶点通过取交集得到144个交互作用靶点,GO功能富集分析确�Objective:Traditional Chinese medicine treatment plan for senile pneumonia was mined by data mining,and the medication rules and action mechanism of TCM on senile pneumonia were analyzed by Traditional Chinese Medicine Inheritance Computer System V3.0 and network pharmacology in order to provide more reference for syndrome differentiation and related research.Methods:By searching with the subject words“senile pneumonia”and“traditional Chinese medicine”in databases such as CNKI,Wanfang database,VIP and so on,the cases of treating senile pneumonia in traditional Chinese medicine were selected and summarized.The traditional Chinese medicine inheritance computer system was used to analyze the medication rules for senile pneumonia,and the pathogenesis and evolution rule of the disease were explored further.All active ingredients and action targets related to the top 15 TCM for treating pneumonia were searched in TCMSP and the bioinformatics analysis tool for molecular mechanism research of TCM medicine.Pneumoniarelated targets were screened by TTD,DrugBank,DisGeNET,GeneCards databases.A“drug-ingredient-target-disease”network was built.The STRING platform was used to build a“medicine-disease target”interaction network.OmicShare cloud platform was used to perform GO enrichment analysis and KEGG pathway enrichment analysis on target proteins.Autodock Vina software was used for a molecular docking analysis.Results:A total of 519 related studies were retrieved in the databases,and there were 81 studies after screening,involving 42 ancient TCM prescriptions on senile pneumonia.Among the 42 ancient TCM prescriptions,the Maxing Shigan decoction(麻杏石甘汤)(13 times),the Qianjin Weijing decoction(千金苇茎汤)or the Weijing decoction(苇茎汤)(10 times),the Bufei decoction(补肺汤)(9 times),and the Qingjin Huatan decoction(清金化痰汤)(8 times)were used more frequently.There were 25 kinds of self-made prescriptions and 6 kinds of Chinese patent medicines,among which Gancao(licorice),Huangqin(skul
