摘 要:目的:基于网络药理学方法探讨吴茱萸汤异病同治原发性头痛和失眠的作用机制。方法:应用中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)获取吴茱萸汤组成药物的活性成分及靶点信息。应用治疗靶点数据库(Therapeutic Target Database,TTD)、药物银行(Drugbank)、比较毒理基因组数据库(Comparative Toxicogenomics Database,CTD)查询原发性头痛及失眠靶点信息。取成分与疾病交集靶点,建立成分-靶点(Compound-Target,C-T)网络,并导入STRING数据库获取蛋白互作信息,借助Cytoscape 3.7.2实现网络可视化并分析关键靶点。应用Cytoscape 3.7.2插件ClueGO对关键靶点进行基因本体(Gene Ontology,GO)和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析。结果:查询得到吴茱萸汤、原发性头痛、失眠靶点分别为137个、507个、607个。获取交集靶点72个,拓扑得到关键靶点33个,主要有过氧化氢酶(Catalase,CAT)、肿瘤蛋白p53(Tumor Protein p53,TP53)、白细胞介素-6(Interleukin-6,IL-6)、肿瘤坏死因子(Tumor Necrosis Factor,TNF)、一氧化氮合酶3(Nitric Oxide Synthase 3, NOS3)等。富集得到对活性氧的反应、对平滑肌细胞增殖的调节、小分子代谢过程的正调控、对镉离子的反应、神经递质代谢过程等70条生物过程信息,以及白细胞介素-17(Interleukin-17,IL-17)信号通路、TNF信号通路、C型凝集素受体(C-type Lectins Receptor,CLR)信号通路、缺氧诱导因子1(Hypoxaia-Inducibble Factor 1,HIF1)信号通路、5-羟色胺(5-hydroxytryptamine,5-HT)能突触信号通路等59条通路信息。结论:吴茱萸汤可能通过干预炎症反应过程、神经递质释放、血管舒缩功能,并介导免疫应答及能量代谢发挥对原发性头痛及失眠的异病同治作用。Objective:To explore the mechanism of network pharmacology of the Wuzhuyu decoction(WZYD)(吴茱萸汤)in treating primary headache and insomnia based on treating different diseases with the same method.Methods:TCMSP was used to screen the compounds and targets of WZYD.TTD,Drugbank and CTD were used to find targets of primary headache and insomnia.Compounds and disease intersection targets were collected and STRING was used to obtain protein interaction information.Cytoscape 3.7.2 was used to realize network visualization and analyze key targets.Result:There were 137,507 and 607 targets of WZYD in primary headache and insomnia respectively.72 intersection targets were obtained,including 23 hub targets such as CAT,TP53,IL6,TNF,and NOS3.In the GO and KEGG enrichment analysis,there were 70 biological processes such as response to reactive oxygen species,regulation of smooth muscle cell proliferation,positive regulation of small molecule metabolic process,response to cadmium ion,neurotransmitter metabolic process and 59 pathways such as IL 17 signaling pathway,TNF signaling pathway,CLR signaling pathway,HIF1 signaling pathway,5-HT signaling pathway.Conclusion:The mechanism of WZYD in treating primary headache and insomnia based on“treating different diseases with the same method”is related to intervening inflammatory response,neurotransmitter release,vasomotor function,and mediating immune response and energy metabolism.
