摘 要:目的研究光动力疗法(photodynamic therapy,PDT)对脊髓背根神经节(dorsal root ganglion,DRG)神经元Nav1.7表达的影响。方法实验对象为原代培养新生SD大鼠DRG神经元,使用NSE免疫荧光染色鉴定细胞。实验分为空白对照组、单纯光敏剂组、单纯激光组和PDT组4组,光敏剂采用浓度为0.5μg/ml血卟啉单甲醚,激光照射光剂量采用波长为635 nm,功率密度为4mW/cm^(2),能量密度为0.16 J/cm^(2)。各组处理后应用CCK8检测细胞活力,qPCR和Western印迹法检测DRG神经元Nav1.7mRNA和蛋白表达量。结果原代培养的DRG神经元纯度达到90%以上。单纯光敏剂组细胞活力与空白对照组比较,差异无统计学意义(P>0.05);单纯激光组和PDT组细胞活力明显降低(P<0.05)。单纯激光组和PDT组Nav1.7 mRNA和蛋白表达量相对于空白对照组均明显升高(P<0.05),并且单纯激光组水平高于PDT组。结论本研究结果发现激光和PDT均可促进DRG神经元Nav1.7表达,其表达增加可能与临床治疗中产生的疼痛有关。Objective To study the effect of photodynamic therapy(PDT)on the expression of Nav1.7 in spinal dorsal root ganglion(DRG)neurons.Methods DRG neurons of newborn SD rats were cultured primarily.The cells were identified with NSE immunofluorescence staining.DRG neurons were divided into the blank control group,simple photosensitizer group,simple laser group and PDT group.The concentration of photosensitizer(hematoporphyrin monomethyl ether)was 0.5μg/ml.The fluence rate of illumination(635 nm semiconductor laser)was 4 mW/cm^(2),and the light dose was 0.16 J/cm^(2).CCK8 was used to detect cell viability.qPCR and Western blot were used to detect the expression of Nav1.7 mRNA and protein in DRG neurons.Results The purity of primary cultured DRG neurons was over 90%.As to cell viability,there was no significant difference between the simple photosensitizer group and the blank control group(P>0.05).However,the cell viability of the simple laser group and the PDT group were significantly reduced(P<0.05).The expression of Nav1.7 mRNA and protein in the simple laser group and the PDT group was significantly higher than that in the blank control group and the expression level of the simple laser group was higher than that in the PDT group(P<0.05).Conclusions Both laser and PDT will promote the expression of Nav1.7 in DRG neurons,which may be related to the pain produced in clinical treatment.This study provides a research basis for treating the pain caused by laser PDT.
