摘  要:目的 探讨在癌症基因芯片GEO数据库、Oncomine数据库、生存分析数据库(Kaplan-Meier Plotter)和蛋白-蛋白相互作用数据库(String)平台中,UBQLN4基因在乳腺癌中的高低表达及临床用药意义。方法 分别对GEO、Oncomine、Kaplan-Meier Plotter、String进行UBQLN4乳腺癌基因表达情况进行深度数据挖掘,分别比较不同类型肿瘤VS正常对应组织、不同种类乳腺癌组织VS正常乳腺组织中基因表达是否存在差异,这些差异基因中,是否存在共表达基因。根据UBQLN4基因的高低表达情况绘制生存曲线,进行UBQLN4基因与乳腺癌患者预后关系分析。并分析UBQLN4蛋白共表达网络,推测UBQLN4可能的相互作用蛋白及其生物学功能,了解紫杉醇对UBQLN4在乳腺癌中的敏感性。结果 检索GEO在GSE4107、GSE5563和GSE27447等3个数据集取交集,得到在乳腺癌组织高表达基因UBQLN4。检索Oncomine数据库发现,在乳腺癌中有3个数据集均显示乳腺癌中UBQLN4高表达,UBQLN4 mRNA共表达显示,UBQLN4与SSR2、ARHGEF2、RXFP4、KIAA0907、RIT1、SYT11、GON4L、ROBLD3等63个基因的mRNA存在着共表达情况;在String蛋白互作的数据库中,对UBQLN4蛋白共表达进行分析,结果显示UBQLN4与RPS27、MRPL24、SLC25A44、LAMTOR2、HAX1等蛋白具有相互作用关系。生存分析显示,UBQLN4mRNA在导管乳腺癌及对照乳腺组织中表达中位生存期分别为553d和517d,差异比较无统计学意义(HR=1.132,95%CI:0.598-2.142,p=0.7037)。UBQLN4mRNA在浸润性导管乳腺癌及对照乳腺组织中表达中位生存期分别为1329d和1466d,差异无统计学意义(HR=0.6711,95%CI:0.3061-1.471,p=0.2866)。应用String数据富集UBQLN4互作作用的可能相关蛋白RPS27、SSR2、RAB13、RAB25、SMG5,发现与UBQLN4相互作用的蛋白有5个,分别为作用评分均大于0.904,5个关联蛋白均属于蛋白编码基因,是核糖体结构成份。且SSR2蛋白也被发现了在各种癌症中存在高表达。在乳腺癌和结肠癌中,�Objective To explore the expression and clinical significance of UBQLN4 gene in breast cancer in the geo database, Oncomine database, survival analysis database(Kaplan-Meier Plotter) and protein-protein interaction database(String) platform.Method In-depth data were carried out on GEO, Oncomine, Kaplan-Meier Plotter and String for gene expression of UBQLN4 breast cancer. Compare whether there were differences in gene expression belween normal tissues and different types of tumor and belween different types of breast cancer tissue and normal breast tissue, and whether there were co-expression genes in these differences.Based on the expression of the UBQLN4 gene, the survival curve was plotted and the prognostic relationship between the UBILN4 gene and breast cancer patients was analyzed. The co-expression network of UBQLN4 protein was analyzed and the possible interaction proteins and their biological functions of UBILN4 was speculated. The sensitivity of yew alcohol to UBIQLN4 in breast cancer was learled about.Result The geo-expression gene UBIQLN4 was obtained by retrieving three data sets, GSE4107, GSE5563 and GSE27447. A search of oncomine database found that in breast ca-ncer, 3 data sets showed high expression of UBQLN4 in breast cancer, UBQLN4 mRNA co-expression showed that UBQLN4 and SSR2, ARHGEF2, There was a co-expression of mRNA in 63 genes, such as RXFP4, KIAA0907, RIT1, SYT11, GON4 L,ROBLD3, etc. The co-expression of UBIQLN4 protein was analyzed, and the results showed that UBILN4 interacted with RPS27, MRPL24, SLC25 A44, LAMTOR2, HAX1 and other proteins.Survival analysis showed that the medium survival of UBQLN4 mRNA expression in catheterized breast cancer and control breast tissue was 553 days and 517 days, respectively, and the difference was statistically unconscionable(HR=1.132,95%CI:0.598-2.142, P=0.7037). The UBQLN4 mRNA expression mid-survival period in indrent catheterized breast cancer and control breast tissue was 1329 days and 1466 days, respectively, and the difference was statistically
