摘  要:目的探讨恒古骨伤愈合剂(Osteoking)对兔激素性股骨头坏死细胞凋亡的影响及机制。方法选取清洁级健康日本大耳白兔24只,18月龄,体重2000~2500 g,采用随机数字表法将其随机分为空白对照组、模型组及恒古骨伤愈合剂组,8只/组。模型组及恒古骨伤愈合剂组采用马血清加激素的方法建立兔激素性股骨头坏死模型,造模成功后24 h,恒古骨伤愈合剂组行恒古骨伤愈合剂420 mg/kg灌胃治疗,1次/2 d,对照组和模型组予以等量的生理盐水灌胃。给药治疗4周后,评价各组兔股骨头组织细胞凋亡指数,caspase-3活性,及Bax、Bcl-2的蛋白表达量。结果与模型组比较,空白对照组和恒古骨伤愈合剂组细胞凋亡指数均较低(P均<0.05),差异均有统计学意义[(7.6±1.0)%比(58.8±3.7)%,(36.7±4.3)%比(58.8±3.7)%]。模型组caspase-3活性较空白对照组升高,差异有统计学意义(P<0.05);恒古骨伤愈合剂组caspase-3活性较模型组降低,差异有统计学意义(P<0.05)。空白对照组Bax蛋白表达量较模型组低,差异有统计学意义[(0.26±0.05)比(0.91±0.08),P<0.05];恒古骨伤愈合剂组Bax蛋白表达量较模型组低,差异有统计学意义[(0.55±0.06)比(0.91±0.08),P<0.05]。空白对照组Bcl-2蛋白表达量较模型组高,差异有统计学意义[(0.73±0.08)比(0.32±0.06),P<0.05];恒古骨伤愈合剂组Bcl-2蛋白较模型组高,差异有统计学意义[(1.33±0.08)比(0.32±0.06),P<0.05]。结论恒古骨伤愈合剂可能通过抑制细胞凋亡,治疗兔激素性股骨头坏死。Objective To explore the effects and mechanisms of Osteoking on apoptosis in hormone-induced femoral head necrosis in the rabbit model.Methods A total of 24 healthy Japanese big ear white rabbits,clean grade,aged 18 months old,weighing 2000-2500 g,were divided into blank control group,model group and Osteoking group by random number table method,8 rabbits in each group.The rabbit model of hormone-induced femoral head necrosis was established by the method of horse serum plus hormones.Then rabbits were received 420 mg/kg Osteoking intragastrically in Osteoking group 24 hours later,1 time/2 d,while rabbits were received the same amount of normal saline intragastrically in the other two groups.After 4 weeks of the treatment,the apoptosis index,caspase-3 activity,and protein expression of Bax and Bcl-2 were detected in all groups.Results Compared with the model group,the apoptosis index was significantly low in the blank control group and Osteoking group[(7.6±1.0)%vs.(58.8±3.7)%,(36.7±4.3)%vs.(58.8±3.7)%,all P<0.05].Compared with the blank control group,the activity of caspase-3 was significantly increased in the model group(P<0.05);compared with the model group,the activity of caspase-3 was significantly decreased in the Osteoking group(P<0.05).The expression of Bax protein was significantly lower in the blank control group and Osteoking group than that in the model group[(0.26±0.05)vs.(0.91±0.08),(0.55±0.06)vs.(0.91±0.08),all P<0.05].The expression of Bcl-2 protein was significantly higher in the blank control group and Osteoking group than that in the model group[(0.73±0.08)vs.(0.32±0.06),(1.33±0.08)vs.(0.32±0.06),all P<0.05].Conclusions Osteoking has a therapeutic effect on hormone-induced femoral head necrosis in rabbits by inhibiting the apoptosis.
