摘  要:目的:观察电针“太阳”透“百会”对戊四氮(PTZ)致痫大鼠行为学、海马内pCREB及NSE表达情况,探讨其抗痫的机制。方法:采用随机分组将32只大鼠分成空白组、模型组、西药组及电针透穴组,每组各8只。除空白组外,采用戊四氮(PTZ)腹腔注射将其余24只大鼠制备为癫痫模型。记录各组大鼠一般状态、行为学改变等情况,免疫组化法及Western-blot检测海马组织pCREB及NSE表达变化情况。结果:模型组、西药组及电针透穴组与空白组比较,体质量明显降低;与空白组比较,模型组大鼠站立及进入旷场中心区探索次数减少,理毛等焦虑行为次数增多,差异具有统计学意义(P<0.05),电针透穴组大鼠站立、进入旷场中心区较模型组增加趋势,理毛等焦虑行为有减少趋势;模型组大鼠海马组织的pCREB及NSE阳性细胞数及细胞核蛋白浓度明显升高,差异具有统计学意义(P<0.05);与模型组比较,西药组、电针透穴组大鼠的体质量均明显增加、痫性发作潜伏期延长和高等级痫性发作减少,差异具有统计学意义(P<0.05);与模型组比较,电针透穴组pCREB及NSE阳性细胞数及细胞核蛋白浓度明显降低,差异具有统计学意义(P<0.05)。结论:头部电针透穴可改善PTZ致痫大鼠一般状态,减轻痫性发作,延长发作潜伏期,改善认知障碍及焦虑行为,可降低海马组织pCREB及NSE表达,其抗痫机制可能通过减轻神经元损伤实现。Objective:To observe the effects of electro-needling Taiyang(EX-HN5)through Baihui(DU20)on behavior,expressions of pCREB and NSE in hippocampus of pentylenetetrazol(PTZ)-induced epileptic rats,and to explore its anti-epileptic mechanism.Methods:32 rats were randomly divided into the blank group,the model group,the western medication group and the electro-acupuncture(EA)point penetration group,with 8 rats in each group.Except the blank group,the other 24 rats were injected intraperitoneally with PTZ to prepare epilepsy model.The general state and behavioral changes of rats were recorded in each group.The expressions of pCREB and NSE in hippocampus were detected by immunohistochemistry and Western blot.Results:Compared to those in the blank group,the body mass was decreased in the model group,the western medication group and the EA point penetration group.Compared to those in the blank group,the times of standing and entering the central area of the open field were reduced,and the times of anxiety behaviors,such as hair grooming,significantly increased in the model group(P<0.05);the times of standing and entering the central area of the open field increased and the anxiety behaviors,such as hair grooming,decreased in the EA point penetration group compared to those in the model group.The number of pCREB and NSE positive cells and the concentration of nuclear protein in the hippocampus increased significantly in the model group(P<0.05).Compared to those in the model group,the body mass of the rats increased significantly,the latency of epileptic attack prolonged and the high-grade attack decreased in the western medication group and in the EA point penetration group(P<0.05).Compared to those in the model group,the number of pCREB and NSE positive cells and the concentration of nuclear protein decreased significantly in the EA point penetration group(P<0.05).Conclusion:Scalp EA point penetration can improve the general state of PTZ-induced epileptic rats,reduce epileptic seizures,prolong onset latency,improve cogniti
