摘 要:目的探讨外源性硫化氢对高脂饮食诱导的肥胖小鼠体重和脂代谢的影响。方法60只小鼠给予高脂饲料喂养8周,建立饮食致胖模型,然后随机均分为模型对照组(B组)、硫化氢钠50μmol·kg^(-1)·d^(-1)处理组(C1组)和硫化氢钠100μmol·kg^(-1)·d^(-1)处理组(C2组);另选取20只小鼠给予低脂饲料喂养,作为正常对照组(A组)。干预2、4、6、8周后,分别记录各组小鼠体重和附睾脂肪组织重量,采用酶比色法检测血清TC和TG水平,HE染色观察肝脏组织和附睾脂肪组织细胞形态变化。结果B组小鼠体重、附睾脂肪组织重量以及血清TC和TG水平均较A组增加(P<0.05)。与B组相比,C2组小鼠肝脏脂肪变性加重,附睾脂肪组织细胞中脂肪堆积增加;C1组干预8周后肝脏脂肪变性程度较B组改善,但附睾脂肪组织细胞中脂肪堆积无明显变化。C1组和C2组干预2、8周后血清TC水平高于B组(P<0.05),干预4周后血清TG水平低于B组(P<0.05);而C2组干预2、6、8周后血清TG水平高于B组(P<0.05)。结论低浓度硫化氢对饮食致胖小鼠肝脏脂肪变性起保护作用,而高浓度硫化氢可能加重肝脏脂肪变性,并增加附睾脂肪组织细胞中的脂肪堆积。Objective To investigate the effect of exogenous hydrogen sulfide on the body weight and lipid metabolism in high fat diet-induced obesity mice.Methods The diet-induced obesity model was established by feeding high fat diet for eight weeks in mice,and then divided into three groups of B(model control),C1(treated with NaSH 50μmol·kg^(-1)·d^(-1))and C2(treated with NaSH 100μmol·kg^(-1)·d^(-1)).Another 20 mice were fed with low fat diet as the normal controls(group A).The body weight and weight of epididymal adipose tissues were recorded 2,4,6 and 8 weeks after intervention.Serum levels of TC and TG were detected by enzyme colorimetry.The changes of cellular morphology of liver tissues and epididymal adipose tissues were observed by HE staining.ResultsThe body weight,weight of epididymal adipose tissues and serum levels of TC and TG in group B were higher than those in group A(P<0.05).Compared with group B,the hepatic steatosis was aggravated and the fat accumulation in epididymal adipose tissue cells was increased in group C2,while the degree of hepatic steatosis after 8 weeks was improved and the fat accumulation in epididymal adipose tissue cells was not significantly changed in group C1.Serum levels of TC after 2 and 8 weeks in groups of C1 and C2 were higher,while which of serum TG after 4 weeks were lower than those in group B(P<0.05).Besides,serum levels of TG after 2,6 and 8 weeks in group C2 were higher than those in group B(P<0.05).Conclusion The lower concentration of hydrogen sulfide has a protective effect on the hepatic steatosis in diet-induced obesity mice,while the higher concentration of hydrogen sulfide may aggravate the hepatic steatosis and increase the fat accumulation in epididymal adipose tissue cells.
