摘 要:以蓝桉种子为试材,采用3因素5水平的均匀试验设计,以MS培养基为基础培养基,并在其中添加不同浓度的6-BA和NAA,对蓝桉种子进行无菌培养,通过对不同处理组合下测定指标的描述统计分析、方差分析和回归分析,探讨琼脂用量和外源激素(6-BA和NAA)浓度对蓝桉实生瓶苗培育的影响效应,构建蓝桉实生瓶苗培育的技术体系,为蓝桉组培技术体系的构建提供充足且理想的外植体材料来源。结果表明,较高浓度的琼脂用量(6.8~8.8 g/L)、一定浓度的NAA(0、0.4、0.8 mg/L)及6-BA(0、0.2、0.8 mg/L)配合使用,可促进蓝桉种子萌发和幼苗生长;方差分析和多重比较发现,既利于蓝桉种子萌发又利于幼苗生长的试验最优处理组合为A_(4)B_(3)C_(1)(琼脂7.8 g/L,NAA 0.4 mg/L,6-BA 0 mg/L);通过回归分析,发芽率、苗高、地径、主根长和鲜重等5个测定指标均求得其可信的回归方程,对回归方程分析认为,理论最优处理组合为A_(5)B_(5)C_(1)(琼脂8.8 g/L,NAA 0.8 mg/L,6-BA 0 mg/L);综合考虑处理措施对蓝桉种子萌发和幼苗生长的促进效果以及处理措施的经济节约性,推荐试验最优处理组合A_(4)B_(3)C_(1)(琼脂7.8 g/L,NAA 0.4 mg/L,6-BA 0 mg/L)为蓝桉实生瓶苗培育的琼脂用量和外源激素用量组合。In this paper,seeds sterile culture of Eucalyptus globulus had been done via the uniform experimental design of three factors at five levels,in which MS medium was used as the basic medium,and different concentrations of agar,6-BA and NAA were added into MS medium.The effects of agar,6-BA and NAA concentrations on the seeds and seedlings sterile culture of E.globulus were researched via descriptive statistical analysis,variance analysis and regression analysis of the indicators with different treatment combinations,then the technical system for seeds and seedlings sterile culture of E.globulus was established for providing sufficient and ideal explants for tissue culture of E.globulus.The results showed that seed germination and seedling growth of E.globulus could be promoted under certain test conditions,namely the high concentration of agar(6.8-8.8 g/L),acertain concentration of NAA(0.0,0.4,0.8 mg/L)and 6-BA(0.0,0.2,0.8 mg/L),according to the descriptive statistical analysis.The optimal experimental treatment combination for both seed germination and seedling growth of E.globulus was A_(4)B_(3)C_(1)(agar 7.8 g/L,NAA 0.4 mg/L,and 6-BA 0.0 mg/L)with Anova and multiple comparison.By the regression analysis,the reliable regression equation was obtained for the five indexes of germination rate,seedling height,ground diameter,primary root length and fresh weight,respectively.Sequencely,the optimally theoretical treatment combination for both seed germination and seedling growth of E.globulus was A_(5)B_(5)C_(1)(agar 8.8 g/L,NAA 0.8 mg/L,and 6-BA 0.0 mg/L)with the analysis of the regression equations.In conclusion,considering the effects of the treatment combinations on seed germination and seedling growth of E.globulus as well as the economic costs of the treatment combinations,the optimal experimental treatment combination(A_(4)B_(3)C_(1))was recommended for guiding the seeds and seedlings sterile culture of E.globulus.
