摘  要:目的探讨体外受精-胚胎移植(IVF-ET)后新鲜胚胎移植(ET)与冷冻胚胎移植(FET)不同移植周期对胎盘中印迹基因PEG10和L3MBTL1表达的影响。方法收集2019年6月至2021年3月在郑州大学第三附属医院行IVF-ET患者分娩的胎盘。根据患者移植周期不同将收集到的胎盘分为两组新鲜胚胎移植组(ET组,16例)与冷冻胚胎移植组(FET组,16例)。比较两组患者的新生儿出生体重,采用实时荧光定量聚合酶链反应、蛋白质印迹法和免疫组化法检测胎盘中印记基因PEG10和L3MBTLI的表达。结果FET组与ET组的孕妇年龄、孕周、孕前BMI、孕期增重、胎盘重量及新生儿出生体重方面比较,差异均无统计学意义(P>0.05)。FET组的新生儿出生体重较ET组有增加趋势(3013.75±388.928)g vs(2871.25±459.273)g。FET组胎盘组织中PEG10 mRNA和蛋白表达4.554±1.992、106.0965±21.6540明显高于ET组1.255±0.4781、77.0741±24.9101,差异有统计学意义(P<0.000,P=0.002),而两组间L3MBTL1表达无显著差异(P>0.05)。结论FET患者子宫内膜更接近生理状态,胎盘中PEG10表达较ET患者增加,进而促进胎儿发育。Objective:To investigate the influences of different cycles of fresh embryo transfer and frozen embryo transfer on the expressions of PEG10 and L3MBTL1 in placenta of IVF-ET.Methods:We collected the placentas delivered by fresh ET and FET in the Reproductive Center of the Third Affiliated Hospital of Zhengzhou University from June 2019 to March 2021.According to the period of embryo transfer,the patients were divided into fresh embryo transfer group(ET group,n=16)and frozen embryo transfer group(FET group,n=16).The real-time quantitative PCR,Western blot and immunohistochemistry were used to detect the expressions of PEG10 and L3MBTLI.Results:There were no significant differences in the women's maternal age,gestational age,BMI before pregnancy,weight gain during pregnancy and placental weight between two groups.The mean neonatal birthweight had a tendency for increasing in FET group.The expression of PEG10 mRNA and protein in FET group was significantly higher than that in fresh ET group(4.554±1.992 vs 1.255±0.478,P<0.000;106.0965±21.6540 vs 77.0741±24.9101,P=0.002),but L3MBTL1 had no significant difference between two groups(P>0.05).Conclusion:The endometrium of patients with FET cycle is closer to the physiological state,and the expression of PEG10 in placenta is higher than that in ET patients,which promotes fetal development.
