摘  要:目的探讨孕中期热暴露后子宫缺血对子代大鼠宫内生长发育的影响。方法36只健康清洁级SD大鼠受孕后随机分为热暴露组(Ⅰ组)、子宫缺血组(Ⅱ组)和热暴露后子宫缺血组(Ⅲ组)。热暴露组妊娠第8~13天置于(35±1℃)环境中进行高温暴露,孕期其余时间温度控制在(23±1)℃;子宫缺血组孕期全程温度控制在(23±1℃),妊娠第14天行双侧子宫动静脉部分结扎;热暴露后子宫缺血组妊娠第8~13天置于(35±1℃)环境中进行高温暴露,孕期其余时间在温度控制在(23±1)℃,妊娠第14天行双侧子宫动静脉部分结扎。测量孕鼠不同妊娠时段的体重变化,检测仔鼠的数量、体重、身长、尾长、外观、活/死胎数、胎盘重量、母体循环HSP70表达及胎盘组织HSP70、Bax、Bcl-2等蛋白表达。结果热暴露后子宫缺血组孕鼠孕期体重增加值低于热暴露组和子宫缺血组(均P<0.05);热暴露后子宫缺血组的胎死率高于热暴露组和子宫缺血组(均P<0.05),窝活胎数低于热暴露组和子宫缺血组(均P<0.05);热暴露后子宫缺血组的胎盘重量低于热暴露组和子宫缺血组(均P<0.05);热暴露后子宫缺血组孕鼠血浆HSP70水平高于热暴露组和子宫缺血组(均P<0.05),但3组胎盘组织HSP70表达比较差异无统计学意义(均P>0.05)。3组胎盘组织Bax与Bcl-2比值(Bax/Bcl-2)热暴露后子宫缺血组>子宫缺血组>热暴露组,组间比较差异均有统计学意义(均P<0.05)。结论热暴露后子宫缺血对大鼠子代宫内发育及胎盘发育有不利影响,增加胎死率,加速胎盘滋养细胞凋亡。Objective:To investigate the effects of uterine ischemia after heat exposure in the second trimester of pregnancy on intrauterine growth of rat offspring.Methods:36 healthy and clean pregnant SD rats were randomly divided into heat exposure group(groupⅠ),uterine ischemia group(GroupⅡ)and uterine ischemia group after heat exposure(GroupⅢ).GroupⅠwas exposed to high temperature at(35±1)℃from the 8th to 13th day of pregnancy,and the rest of the time was kept at(23±1)℃.In groupⅡ,the whole pregnancy temperature was kept at(23±1)℃,and partial ligation of bilateral uterine arteries and veins was performed on the 14th day of pregnancy.GroupⅢwas exposed to high temperature at(35±1)℃from the 8th to 13th day,and was raised at(23±1)℃for the rest of pregnancy.Bilateral uterine arteries and veins were partially ligated on the 14th day of pregnancy.Measurement of pregnant rats pregnancy at different times of the weight value added.In late pregnancy,the number,weight,body length,tail length and appearance of fetal rats were detected.At the same time,the quality of placenta,the expression of HSP70 in maternal circulation and the expressions of HSP70,Bax,Bcl-2 in placenta were detected.Results:The weight gain of pregnant rats in groupⅢwas lower than that in groupsⅠandⅡ(P<0.05).The fetal death rate in groupⅢwas higher than that in groupⅠand groupⅡ(P<0.05),and the number of live births was lower than that in groupⅠand groupⅡ(P<0.05).The placental weight of groupⅢwas lower than that in groupⅠand groupⅡ(P<0.05).The circulating HSP70 level in groupⅢwas higher than that in groupⅠand groupⅡ(P<0.05),but there was no significant difference in the expression of HSP70 in placenta among the three groups(P>0.05).The ratio of Bax and Bcl-2(Bax/Bcl-2)in placental tissue of three groups:GroupⅢ>groupⅡ>groupⅠ(P<0.05).Conclusion:Uterine ischemia after heat exposure will bring adverse effects on the development of placenta and offspring,increase the fetal death rate,and accelerate the apopto
