籽粒苋不同器官内源激素含量比较研究

摘　　要：文章对影响植物内源激素提取的材料预处理、细胞破碎和提取方式等关键因素进行筛选,通过ELISA检测,获得优化实验方案后测定籽粒苋不同器官的内源激素含量.结果显示:取籽粒苋鲜材料、石英砂研磨破碎细胞和制备粗提液,ELISA检测效果好;籽粒苋14天幼苗胚根、胚轴、子叶和茎尖四种器官中,ZT,GA,GH,CTK,ABA含量最高的是茎尖,ZT,GH,CTK,ABA含量最低的是胚轴,GA含量最低的是胚根.研究结果可为籽粒苋组织培养提供参考依据.This article screens the key factors that affect the material pretreatment,cell disruption,and extraction methods of plant endogenous hormone extraction,and obtains an optimized experimental program by ELISA detection effect. The method was used to determine the content of endogenous hormones in different organs of Amaranthus hypochondriacus L.. The results showed that the fresh material of Amaranthus hypochondriacus L. was taken,and the broken cells were grinded with sands to prepare a crude extract. The effect of ELISA was good. The 14-day-old organs of grain amaranth seedlings had radicle,hypocotyl,cotyledon and shoot tip.The highest ZT,GA,GH,CTK and ABA content is the shoot tip;the lowest ZT,GH,CTK and ABA content is the hypocotyl,the lowest GA content is the radicle.This result can provide a reference for the tissue culture of Amaranthus hypochondriacus L..
