摘 要:糖尿病肾脏病(DKD)是糖尿病患者最主要的微血管并发症之一,其发病机制主要与代谢紊乱、血流动力学异常、氧化应激等有关,炎性反应贯穿于DKD全过程。细胞焦亡(pyroptosis)是新发现的一种细胞死亡形式,由NOD样受体蛋白3(NLRP3)炎症小体激活,依赖半胱氨酸蛋白水解酶(caspase)-1/4/5/11,通过Gasdermin D(GSDMD)介导释放炎性因子,发生炎性反应。细胞焦亡与DKD的发病机制密切相关,本文对细胞焦亡在DKD发病中的作用、分子调控机制以及相关干预研究进行综述。Diabetic kidney disease(DKD)is one of the most important microvascular complications of diabetes.The pathogenesis of DKD is mainly related to metabolic disorders,hemodynamics change,oxidative stress and so on.Inflammation is involved in the whole progression of DKD.Pyroptosis,a new kind of programmed cell death,is activated by NOD-like receptor pyrin domain-containing 3(NLRP3)inflammasome and depends on caspase-1/4/5/11.Finally,inflammatory cytokines are released through Gasdermin D(GSDMD),leading to inflammatory response.Pyroptosis is closely related to the pathogenesis of DKD.In this paper,we summary the current understanding of pyroptosis and its role in the pathogenesis,molecular mechanism,and related interventions of DKD.
