摘 要:目的:应用乙醇提取联合超声波辅助技术优化黄葵花总黄酮提取工艺流程。方法:选用黄葵花为原料,应用乙醇提取联合超声波辅助技术提取总黄酮,采用改良酶标仪比色法测定总黄酮含量,通过单因素实验法与正交试验相结合的方法,优化黄葵花总黄酮提取工艺。结果:正交试验显示,醇提联合超声的最佳提取工艺为样筛孔径90μm,料液比1∶100 g/ml,乙醇浓度60%,浸泡时间2 h,提取次数3次,提取温度60℃,提取时间20 min。该工艺测定黄葵花总黄酮平均含量为6.807%,标准偏差在2%以内,平均回收率为98.73%。结果提示样筛孔径、浸泡时长、提取次数、提取时间可对提取率产生明显影响。结论:优化的黄葵花总黄酮提取工艺重复性好、稳定、高效,可用于大批量生产。Objective:To optimize the process of extracting total flavonoids from Abelmoschus manihot by appling ethanol extraction combined with ultrasonic auxiliary technology.Methods:Abelmoschus manihot(L.)flower was chosen as the raw material,and ultrasonic-assisted ethanol extraction was applied to extracting total flavonoids.Then,the total flavonoid content of Abelmoschus manihot was determined by modified microplate reader visible spectrophotometry,and the extraction process was optimized by the combination of single-factor experiment and orthogonal test.Results:Orthogonal test showed that the optimal extraction process of ethanol extraction combined with ultrasound was obtained as the following:Sample sieve aperture was 90μm,ratio of raw material to extract was 1∶100 g/ml,ethanol concentration was 60%,soaking time was 2 h,extraction times were 3 times,extraction temperature was 60℃,and extraction duration was 20 min.The average content and relative standard deviation(RSD)of the total flavone determination was 6.807%,2%,and the average recovery efficiency was 98.73%.The results suggest that sample sieve aperture,soaking time,extraction times,and extraction duration exerted the significant effects on the extraction rate.Conclusion:The optimized extraction process of total flavonoids from Abelmoschus manihot flower has good repeatability,stability and efficiency,which can be used for mass production.
