小四五颗粒调节狼疮肾炎小鼠的免疫功能及机制

摘　　要：目的:探讨小四五颗粒对MRL/MpJ-Faslpr小鼠免疫功能的影响及其机制。方法:将MRL/MpJ-Faslpr小鼠分为治疗组和狼疮组,治疗组给予0.2 ml/d小四五灌胃,狼疮组及对照组均给予等量生理盐水灌胃,给药6周,收集小鼠尿液检测尿蛋白含量;采用ELISA检测小鼠血清中IL-4、IL-10、IFN-γ含量;流式细胞数检测各组小鼠脾脏组织中的Th1/Th2细胞比例。结果:LN小鼠与对照小鼠相比,尿蛋白含量显著增高,治疗组小鼠与狼疮组小鼠相比,尿蛋白含量显著下降;小四五颗粒能显著降低MRL/MpJ-Faslpr小鼠血清IL-4、IL-10含量,增加IFN-γ含量,同时能减少Th2细胞,增加Th1细胞,显著升高Th1/Th2细胞比值。结论:小四五颗粒能上调MRL/MpJ-Faslpr小鼠体内Th1/Th2细胞比值,同时减少尿蛋白含量,减缓肾损伤,进而发挥治疗狼疮肾炎的作用。Objective:To investigate the effects of Xiaosiwu granule on immune function of MRL/MpJ-Faslpr mice and its mechanism.Methods:MRL/MpJ-Faslpr mice were divided into the treatment group and lupus group.The treatment group was given 0.2 ml xiaosiwu granule intragastric administration every day,while the lupus group and control group were given the same amount of physiological saline gavage.After 6 weeks of administration,the urine of mice was collected to detect the content of urinary protein;IL-4,IL-10 and IFN-γcontent in mouse serum were detected by ELISA;the proportion of Th1/Th2 cells in spleen tissue of mice in each group was detected by flow cytometry.Results:Compared with mice in the control group,the content of urinary protein in LN mice increased more significantly,and the content of urinary protein in treatment group decreased more significantly than that in lupus group.Xiaosiwu granule could significantly reduce the contents of serum IL-4 and IL-10 and increase IFN-γin MRL/MpJ-Faslpr mice.At the same time,it could reduce Th2 cells,increase Th1 cells and significantly increase the ratio of Th1/Th2 cells.Conclusion:Xiaosiwu granule can up-regulate Th1/Th2 cell ratio in MRL/MpJ-Faslpr mice,reduce urinary protein content,slow down renal injury,and then play an important role in the treatment of lupus nephritis.
