摘 要:【目的】探讨丹酚酸B对慢性肾盂肾炎大鼠肾间质损伤的影响及机制。【方法】将40只SD大鼠分为对照组(生理盐水灌胃)、模型组(生理盐水灌胃)、丹酚酸B组(12.5 mg/kg丹酚酸B灌胃)、激活剂组(12.5 mg/kg丹酚酸B灌胃+2.5 mg/kg Smad3激活剂SRI-011381腹腔注射),每组10只。模型组、丹酚酸B组、激活剂组建立慢性肾盂肾炎大鼠模型,建模次日开始给药,每日1次,连续6周。检测各组大鼠血肌酐、24 h尿蛋白含量、肾组织中α平滑肌肌动蛋白(α-SMA)、Smad3、转化生长因子-β1(TGF-β1)的表达情况。【结果】与对照组比较,模型组24 h尿蛋白含量升高(P<0.05);与对照组比较,模型组肾组织中α-SMA表达量升高(P<0.05);与模型组比较,丹酚酸B组及激活剂组肾组织中α-SMA表达量降低,且丹酚酸B组低于激活剂组(P<0.05)。与对照组比较,模型组肾组织中p-Smad3、TGF-β1、α-SMA蛋白相对表达量升高(P<0.05);与模型组肾组织中p-Smad3、TGF-β1、α-SMA蛋白相对表达量比较,丹酚酸B组及激活剂组均降低,且丹酚酸B组低于激活剂组(P<0.05)。【结论】丹酚酸B可抑制慢性肾盂肾炎大鼠肾间质纤维化,其调控机制可能与抑制Smad3信号通路有关。【Objective】To investigate the effect of salvianolic acid B on renal interstitial injury in rats with chronic pyelonephritis and its mechanism.【Methods】Forty SD rats were divided into control group(normal saline gavage),model group(normal saline gavage),salvianolic acid B group(12.5 mg/kg salvianolic acid B gavage)and activator group(12.5 mg/kg salvianolic acid B gavage+2.5 mg/kg Smad3 activator SRI-011381 intraperitoneal injection),with 10 rats in each group.The rat model of chronic pyelonephritis was established in the model group,the salvianolic acid B group and the activator group.The drug was administered once a day for 6 weeks from the next day.Expression of the contents of serum creatinine,24-hour urinary protein and renal tissue,αSmooth muscle actin(α-SMA),Smad3,transforming growth factor-β1(TGF-β1)were measured.【Results】Compared with the control group,the 24-hour urinary protein content in the model group increased(P<0.05);Compared with the control group,the expression ofα-SMA of the renal tissue in the salvianolic acid group B and the activator group decreased(P<0.05);Compared with the control group,the expression ofα-SMA in the salvianolic acid group B was lower than that in the activator group(P<0.05).Compared with the control group,the relative expression of P-smad3,TGF-β1 andα-SMA protein of renal tissue in the model group increased(P<0.05);Compared with the relative expression of p-Smad3,α-SMA protein and TGF-β1 in kidney tissue of the model group,those of the salvianolic acid group B and the activator group decreased,and those of the salvianolic acid group B was lower than those in the activator group(P<0.05).【Conclusion】Salvianolic acid B can inhibit renal interstitial fibrosis in rats with chronic pyelonephritis,and its regulatory mechanism may be related to the inhibition of Smad3 signal pathway.
