摘  要:目的采用高效液相色谱法测定大尺度不同产地五指毛桃中补骨脂素的含量,进行五指毛桃的质量评价。方法采用HPLC法,色谱柱:Agilent 5 HC-C_(18)(4.6mm×250mm,5μm);流动相:乙腈-水(35∶64),流速为1.0ml/min;检测波长为254nm;柱温为30℃。结果补骨脂素在20~60μg/ml范围内线性关系良好,平均回收率为93.69%(RSD=0.99%)。不同产地的五指毛桃补骨脂素含量存在一定差异(0.018%~0.073%),道地产区平均含量0.048%,非道地产区平均含量0.023%。结论不同产地的五指毛桃的质量存在一定差异,道地产区优于非道地产区。研究可为五指毛桃质量标准的制定和临床用药提供参考依据。Objective To establish the High Performance Liquid Chromatography(HPLC)method for the determination of psoralen content in Radix Fici Hirtae from large-scale eight different producing areas and conduct evaluation on the quality.Methods HPLC with Agilent 5 HC-C_(18)(4.6 mm×250 mm,5μm)column was used.Acetonitrile-water(35:64)was used as the mobile phase and the volume flow rate was 1.0 ml/min.Detection wave length was 254 nm and The column temperature was 25℃.Results The linear relationship is good in the range of 20~60μg/ml and the average recovery rate is 93.69%(RSD=0.99%).The psoralen content in Radix Fici Hirtae from different producing area is different(0.018%to 0.073%),the average psoralen content of Radix Fici Hirtae from genuine producing areas is 0.048,while the average psoralen content o from non-genuine areas is 0.023%.Conclusion The quality of Radix Fici Hirtae from different producing areas have some differences,the quality of Radix Fici Hirtae from genuine producing areas is better than that from non-genuine areas.This study can be used as reference for the establishment of quality standard of Radix Fici Hirtae and the use in clinical practice.
